脂肪组织缺氧与胰岛素抵抗的相关性研究

摘    要:目的:检测人脂肪组织缺氧并探讨其与胰岛素抵抗的相关性。方法:收集16名非肥胖(BMI<28 kg/m2)与18名肥胖(BMI>28 kg/m2)女性腹部脂肪组织,Real-time PCR测定缺氧诱导因子-1α(HIF-1α)mRNA的表达,Western Blot检测HIF-1α蛋白的表达。检测空腹外周血血糖(FBG)和胰岛素(FINS),计算HOMA指数评估胰岛素抵抗。检测HIF-1α表达与HOMA指数的相关性。结果:与非肥胖组相比,肥胖组腹部脂肪组织HIF-1αmRNA和蛋白的表达均显著升高(P<0.028,P<0.005);且肥胖组HOMA指数显著高于非肥胖组(P<0.01);HIF-1αmRNA和蛋白表达与HOMA指数之间存在相关性(r=0.360,P<0.05;r=0.343,P<0.05)。结论:肥胖患者脂肪组织HIF-1α的表达升高并存在周身胰岛素抵抗,且两者具有相关性。

Abstract:Objective: To detect hypoxia in human adipose tissues and study the correlation between hypoxia and insulin resistance.Methods: Abdominal adipose tissues of 16 non-obese females(BMI<28 kg/m 2) and 18 obese females(BMI>28 kg/m 2) were collected by surgery.The mRNA expression of hypoxia induced factor-1α(HIF-1α) in adipose tissue was detected by real-time PCR.The expression of HIF-1α protein was determined by Western Blot.The concentrations of FBG and FINS in peripheral blood were measured and the HOMA index was calculated to determine whole body insulin resistance.The correlation between HIF-1α level and HOMA index was determined by correlation analysis.Results: The mRNA and protein expressions of HIF-1α and HOMA index were higher in obese adipose tissue than those in non-obese subjects(P<0.028,P<0.005,P<0.01,respectively).The mRNA and protein expressions of HIF-1α were correlated to HOMA index(r=0.360,P<0.05;r=0.343,P<0.05,respectively).Conclusion: Obese subjects have hypoxic adipose tissues and systemic insulin resistance.The expression of HIF-1α is correlated to HOMA index.
