NS-398对宫颈癌Hela细胞顺铂敏感性的影响

目的 研究环氧合酶-2（COX-2）特异性抑制剂NS-398对宫颈癌Hela细胞顺铂敏感性的影响.方法 用含NS-398和顺铂的细胞培养液作用于Hela细胞,应用四甲基偶氮唑蓝（MTT）比色法检测作用后的Hela细胞对顺铂敏感性的变化;采用流式细胞仪分别检测经过NS-398和顺铂作用的细胞与顺铂联合单独作用的细胞的COX-2蛋白表达情况.结果 NS-398和顺铂联合作用后的Hela细胞COX-2表达量较顺铂单独作用后的减少,差异有统计学意义（P〈0.05）;在转染48h后,Hela细胞对顺铂的半数抑制量（IC50值）从（10.475±1.713）μg/ml提高到（0.194±0.021）μg/ml,Hela细胞对顺铂的敏感性增加了53.88倍.结论 NS-398能增加宫颈癌Hela细胞对顺铂的敏感性.

NS-398 enhances cytotoxicity of cisplatin in cervical carcinoma Hela cells

Objective To investigate the effect of cyclooxygenase-2 （COX-2） inhibitor NS-398 on the sensitivity of cisplatin in cervical carcinoma Hela cells. Methods The cultured cervical carcinoma Hela cells were treated with cisplatin and 0.05M NS-398 was added in culture media. The COX-2 protein levels were measure by flow cytometry, and the changes of cisplatin sensitivity were examined by methyl thiazolyl tetrazolium （MFI-） assay. Results COX-2 levels in Hela cells were dramatically de- creased at 24, 48 and 72 h after adding NS-398. The sensitivity of Hela cells to cisplatin was increased by 53.88 folds. Conclusion NS-398 can enhance the sensitivity of cervical carcinoma Hela cells to cisplatin, which is associated with the inhibition of COX-2 levels in the cells.
