摘    要:目的 观察血栓调节蛋白(TM)在肺动脉高压模型大鼠表达的变化,以探讨其在肺血管重塑中的作用。方法 利用野百合碱(MCT)诱导大鼠肺高压动物模型,右心导管法测定大鼠肺动脉压力,用双抗体夹心酶联免疫吸附法检测不同时间点大鼠血浆TM水平,采用逆转录定量聚合酶链反应(RT-PCR)检测肺组织中TM mRNA表达水平。结果实验第14天肺动脉压力开始升高,第21天明显升高,以第28天为最高。模型组大鼠血浆TM浓度在第7天时有升高,第14天时明显升高,与对照组有显著差异;第21天开始下降,第28天下降显著,与对照组相比有显著差异。模型组大鼠肺组织中TM mRNA水平在第7天时即开始表达减少,第14天时表达明显减少,以28天时表达最低。结论 血栓调节蛋白在肺动脉高压时表达减少,可能在肺动脉高压发病机制中发挥重要作用,提示肺动脉高压时需要适当抗凝治疗。

Abstract:Objective To evaluate the role of thrombomodulin (TM) in the pulmonary vascular remodeling in pulmonary hypertensive rats. Methods The pulmonary hypertension animal model was induced by monocrotaline (MCT), and the pulmonary arterial pressure was measured by catherization. The pulmonary TM mRNA expression was studied by RT-PCR and plasma levels of TM were determined by enzyme-linked immunosorbent assay.Results The pulmonary arterial pressure increased significantly on the 21st day after injection of MCT and reached at the peak on the 28th day. The plasma levels of TM increased during the first two weeks,and then decreased significantly. The pulmonary TM mRNA expression began to reduce on the 7th day after injection of MCT. TM mRNA expression in the lungs was significantly lower in test rats than that in the control on the 14th day and was the lowest on the 28th day after injection. Conclusion Decreased thrombomodulin expression may reflect in situ thrombosis due to endothelial injury,which might play an important role in the pathogenesis of pulmonary hypertension.Anticoagulation therapy may be required in the pulmonary hypertersives.
