摘    要:目的:探讨当归散对骨折早期局部血管内皮增长因子表达的影响。方法:72只雄性SD大鼠建立开放骨折髓内固定动物模型,分当归散组、生理盐水组。两组分别于术后第3、5、7、14、21d相同时间点每组各取6只处死,取股骨全长标本,观察大体标本骨折愈合情况,并在同一条件下摄正侧位X射线片,在无菌操作下快速取下骨折区组织,RT-PCR检测VEGF mRNA的表达。结果:VEGF mRNA在大鼠骨折第3天表达开始增强,当归散组术后第3天VEGF表达较生理盐水组弱,差异无统计学意义(P〉0.05);第5、7、14、21天当归散组表达较对照组强,第7天VEGF表达到峰值,与生理盐水组比较差异有统计学意义(P〈0.01)。结论:骨折早期当归散促进VEGF表达增强。

Effect of Angelica sinensls powder on activity of vascular endothelial growth factor in early stage of fracture

Abstract:Objective :To investigate the effect of Angelica sinensis powder on the local expression of vascular endothelial growth factor (VEGF) in the early stage of fracture. Methods:Seventy - two male Sprague - Dawley rats were treated to establish a model of intramedullary fixation for open fracture and divided into Angelica sinensis powder group and normal saline group. In each group,6 rats were sacrificed at the same time point at 3,5,7,14, or 21 d after operation. The whole femur specimens were collected ;the fracture healing of gross specimens was evaluated, and anteroposterior and lateral X - ray films were taken under the same condition ; the tissues in fracture area were rapidly taken by aseptic operation, and the mRNA expression of VEGF was measured by RT - PCR. Results:The mRNA expression of VEGF began to increase on the 3rd d after fracture, and the Angelica sinensis powder group had a nonsignificantly lower VEGF expression than the normal saline group (P 〉 0. 05 ). On the 5th ,7th, l4th, and 21st d after fracture, the Angelica sinensis powder group had a significantly higher VEGF expression than the control group, and the VEGF expression reached the peak level on the 7th d,with a significant difference compared with the normal saline group (P 〈 0. 01 ). Conclusion:Angelica sinensis powder can enhance the VEGF exoression in the early stage of fracture.
