心脑通络液对动脉粥样硬化兔平滑肌细胞Bcl-2、Caspase-3的影响

摘    要:目的:探讨心脑通络液对动脉粥样硬化兔平滑肌细胞的干预作用。方法:选取健康雄性新西兰大耳兔40只。随机分成假手术组、模型组、血脂康组、心脑通络液高、低剂量组,对除假手术组外的其余组动物建立腹主动脉粥样硬化模型。造模成功后,各组分别灌胃给药,连续治疗8周,采用SP法测定各组家兔Bcl-2、Caspase-3的阳性表达率。结果:与假手术组比较,模型组Bcl-2含量明显降低,Casapase-3含量明显升高,差异有统计学意义(P〈0.05);与模型组比较,血脂康组和高低剂量组Bcl-2含量均增高、Caspase-3含量均降低(P〈0.05),以高剂量改善最显著(P〈0.05)。结论:心脑通络液可能通过对bcl-2及Caspase-3蛋白表达的调控而降低动脉斑块内过度凋亡的平滑肌细胞,从而抑制粥样斑块的形成。

Effects of Xinnao Tongluo liquid on Bcl - 2 and Caspase - 3 in smooth muscle cells of rabbits with atherosclerosis

Abstract:Objective :To investigate the intervention effects of Xinnao Tongluo liquid on the smooth muscle cells of rabbits with atherosclerosis. Methods : Forty healthy male New Zealand rabbits were randomly divided into sham - operation group, model group, Xuezhikang group, and high- and low - dose Xinnao Tongluo liquid groups. All rabbits except those in sham - operation group were treated to establish a model of abdominal aorta atherosclerosis. After the model was successfully established, each group received drugs by gavage for 8 consecutive weeks. The positive rates of Bcl - 2 and Caspase - 3 expression in smooth muscle cells were determined by SP method. Results : Compared with the sham - operation group, the model group had a significantly reduced Bcl - 2 level and a significant- ly increased Caspase- 3 level(P 〈 0. 05 ). Compared with the model group, the Xuezhikang group and high - and low -dose Xinnao Tongluo liquid group had significandy increased Bcl - 2 levels and significantly reduced Caspase - 3 levels ( P 〈 0. 05 ) , and the high - dose Xinnao Tongluo liquid group showed the most improvement(P 〈 0. 05 ). Conclusion :Xinnao Tongluo liquid can reduce the excessive apoptosis of smooth muscle cells in artery plaques by regulating the protein expression of Bcl - 2 and Caspase - 3, thus inhibiting the formation of atheromatous plaque.
