| 摘 要： | 目的 研究优质的抗人红细胞血型糖蛋白Ａ单克隆抗体（ＭＣＡＢ－ＧＰＡ）因为该抗体是研制快速全血凝集试剂中双功能抗体的关键成份，为在我国研制能诊断多种疾病的快速全血系列诊断试剂创造条件，方法 首先用ＭＮ亚型ＧＰＡ（购于美国Ｓｉｇｍａ公司）免疫小鼠，采用常规杂交瘤细胞技术进行融合和克隆，用ＥＩＡ和血凝方法进行生物特性鉴定，结果 建立１２个ＭＣＡＢ－ＧＰＡ株，１２个抗体株与ＧＰＡ发生特异性反应。但对Ａ，Ｂ

| Abstract: | ObjectiveTo study and prepare monoclonal antibody to glycophorin A of human erythrocyte(MCAB GPA).This antibody is a key reagent in prepration of bispecific antibodies for rapid whole blood immunoassay.MethodsBALB/c mice were injected with GPA antigen.Hybridoma was produced by using traditional techniques.Hybridoma were determined with method of EIA and indirect agglutination(IA).ResultsAll 12 MCAB GPA were reacted with GPA,and they were not autoagglutination with four types of red cells(type A,B,O,AB).Of 12 MCAB GPA,7 antibodies were IgG 1,4 IgG 2a and only one IgG 2b subtypes.7 IgG 1 MCAB GPA can be used in makng bispecific antibody in preparation of rapid whole blood immunoassay.ConclusionThis result is an important foundation for making bispecific antibodies in preparation of rapid whole blood immunoassay. |
