老年2型糖尿病周围神经病变患者外周血调节性T细胞的检测及其临床意义

摘    要:目的:探讨2型糖尿病周围神经病变(DPN)患者外周血中CD4+CD25+Foxp3+调节性T细胞(Treg)的变化及其检测的临床应用。方法:采用流式细胞技术多色荧光分析法,对20例初诊老年DPN患者、20例初诊老年2型糖尿病(T2DM)不伴并发症患者及20例性别年龄匹配的健康对照(CON)者外周血中Treg细胞测定,对两组结果进行统计学分析。结果:与正常对照组比较,DPN组和DM组患者外周血CD4+CD25+Foxp3+ Treg细胞数量均显著减少(P<0.05),且DPN组患者较DM组患者亦显著减少(P<0.05);DPN组患者外周血CD4+CD25+Foxp3+ Treg细胞比例与MNSI评分呈负相关,但无统计学意义,而与MDNS评分则呈显著负相关(r=-0.339,P<0.05)。结论:老年DPN患者中CD4+CD25+Foxp3+ Treg细胞数量减少,可能在DPN的发生发展中起到一定作用。

Abstract:Objective To study the change of CD4+CD25+Foxp3+ regulatory T cell (Treg) in peripheral blood of elderly patients with type 2 diabetic peripheral neuropathy (DPN) and its clinical application. Methods CD4+CD25+Foxp3+ Treg were detected by flow cytometry in peripheral blood of elderly patients with DPN (20 cases), type 2 diabetes mellitus (T2DM, 20 cases) and age-matched healthy donors (CON, 20 cases). Results The proportion of CD4+CD25+Foxp3+ Treg in DPN group and T2DM group were significantly lower than that in healthy donors (P < 0.05), as well as in DPN cases than that in T2DM cases (P < 0.05). The proportion of CD4+CD25+Foxp3+ Treg was significantly negatively correlated with MDNS (r = -0.339, P < 0.05). Conclusion The proportion of CD4+CD25+Foxp3+ Treg is decreased in elderly patients with DPN, which may play a role in the progress of DPN in elderly patients.
