摘    要:目的:探讨同型半胱氨酸(homocysteine,Hcy)对促进血管平滑肌细胞(vascular smooth muscle cells,VSMCs)增殖的影响.方法:常规培养人脐静脉VSMCs,将培养的VSMCs分为0、50、100、200、500及1000 μmol/L Hcy,其中0 μmol/L Hcy组为对照组.当VSM-Cs细胞长满80%瓶底时,各组加入不同浓度的Hcy,继续培养细胞6h、12h、24h、36h、48h、72h后,采用倒置显微镜观察细胞的生长状态,采用MTT法检测细胞的增殖活力.结果:当Hcy刺激VSMCs 12~48h时,VSMCs的细胞数逐渐增加,细胞的活力也持续增加(P<0.05);当Hcy的刺激时间达72h时,高浓度(1000 μmol/L Hcy)组的VSMCs活力的升高趋势有所减缓,细胞有损害表现.结论:高浓度Hcy可直接诱导VSMCs增殖;但随着刺激时间的增加,过高浓度的Hey对VSMCs表现出抑制和损害效应.

Abstract:Objective:To investigate the impact of homocysteine(Hcy) on the proliferation of vascular smooth muscle cells(VSMCs).Methods:Cultured human umbilical VSMCs were divided into 0,50,100,200,500,1000 μmol/L Hcy groups,and the 0 μmol/L Hcy group was the control.After the VSMCs were treated with different concentrations of Hcy for 6h,12h,24h,36h,48h and 72h,the morphology and the viability of VSMCs were detected by MTT and inverted microscope respectively.Results:The number and viability of VSMCs were increased after the VSMCs were stimulated with Hcy from 12h to 48h.But the increase of VSMCs stimulated up to 72h was slowed down in 1000 μmol/L Hcy group.Conclusion:Increased concentration of Hcy could induce proliferation of VSMCs,while severely increased concentration of Hcy could injury VSMCs and inhibit its proliferation with the time of stimulation prolonging.
