大鼠颊黏膜鳞癌细胞系端粒酶活性与其转移相关性的研究

摘    要:目的:检测SD大鼠颊黏膜鳞癌细胞系SDSCC—B与其单克隆细胞系Rca—B细胞的端粒酶活性,分析其对细胞转移能力的影响。方法:采用单细胞克隆培养法筛选细胞系SDSCC—B,建立单克隆来源的细胞系Rca—B;分别检测2个细胞系端粒酶的活性:以Tca8113和Tb细胞系做阳性对照。2个细胞系经裸鼠尾静脉注射后,观察其实验性肺转移情况。检测结果采用SPSS11.0.1软件包分别进行q检验和t检验。结果:端粒酶活性检测结果显示,2个细胞系的端粒酶活性均为强阳性.显著高于大鼠正常颊黏膜细胞,差别均具有统计学意义(P〈0.01);Rca—B端粒酶活性高于SDSCC—B细胞,差别具有统计学意义(P〈0.05);同时。Rca—B细胞实验性肺转移能力较后者高,Rca—B组与SDSCC—B组肺重,体重的比值差别具有统计学意义(P〈0.05)。结论:SD大鼠细胞系SDSCC—B和Rca—B均具有高转移特性,端粒酶活性的表达与转移能力有关,Rca—B的转移能力更强。

Abstract:PURPOSE:To study the relationship of the telomerase activity and the potential of experimental metastasis to the lung between Sprague-Dawley rat buccal squamous cell carcinoma cell line and its purified sub-cell line. METHODS:In vitro inbred line Rca-B was purified from SDSCC-B cell line by using mono-cell cloning method. The telomerase activity of cell lines was analyzed by TRAP-ELISA. The ratios of LW/BW (lung weight/body weight)of nude mice after experimental lung metastasis were used to evaluate the metastatic characters. The results were analyzed by SPSS11.0.1 software package for q test and t test respectively. RESULTS: The results showed that the telomerase activity of the cell lines was significantly higher than normal buccal mucosa cells(P<0.01), telomerase activity of Rca-B cells was significantly higher than that of SDSCC-B cells(P<0.05). The ratios of LW/BW of nude mice of the two cell lines were significantly different(P<0.05). CONCLUSION:Rca-B and SDSCC-B cells were of high metastatic ability, and Rca-B was more easily metastatic and lower differenciated .The expression of telomerase activity was relevant with the metastatic ability of these cells.
