乙醇对雄性大鼠交配行为的影响及其机制研究

摘    要:目的探讨乙醇对雄性大鼠下丘脑神经递质、血中激素水平和交配行为的影响及其机制。方法健康wistar雄性大鼠40只,随机分为4组:对照组、低浓度乙醇组、中浓度乙醇组和高浓度乙醇组。在灌服不同浓度的乙醇后2h,分别应用交配实验、乙醇脱氢酶法、放射免疫分析法、硝酸还原酶法和化学比色法观察乙醇中毒后雄性大鼠交配行为的改变,血清睾酮(Testosterone,T)、下丘脑一氧化氮(NO)、一氧化氮合酶(NOS)含量的变化。结果各实验组大鼠骑跨次数和插入次数与对照组大鼠比较明显减少,差异有显著性(P〈0.05或P〈0.01),乘骑潜伏期及插入潜伏期较对照组大鼠明显延长(P〈0.01);实验组大鼠血清睾酮含量较对照组均下降,下丘脑NO,NOS含量明显降低,差异有显著性(P〈0.01)。结论急性乙醇中毒后,下丘脑神经递质NO含量的降低和血中T水平的降低,可能是乙醇引起大鼠交配行为改变的机制之一。

Abstract:Objective To explore the effect of alcohol on neurotransmitters in hypothalamus, blood serum testosterone and mating behaviors and approach its possible mechanism. Methods Forty healthy male Wistar rats were randomly divided into the control group, low concentration group, middle concentration group and high concentration group. Induced with alcohol solution 2 hours later,the following indexes were detected:①Sexual behavior changes with the mating test. ②Blood serum testosterone changes with radio immunoassay.③Changes of nitric oxide (NO) and nitric oxide synthase(NOS) in hypothalamus by nitrate reductase and chromatometry methods. Results Contrasted with the control group, mounting and intromission times decreased significantly in the experimental groups ( P 〈 0.05 or P 〈 0. 01 ) ,while the mating and intromission latency periods of the experimental groups became prolonged obviously ( P 〈 0.01) ;Blood serum testosterone of experimental groups was decreased;Compared with the control group, NO and NOS in hypothalamus of the experimental groups were significantly lower( P 〈 0.01 ). Conclusion In acute alcohol intoxication, reduction of NO in hypothalamus and blood serum testosterone may be the possible mechanism which makes the mating behaviors change.
