高效液相色谱法测定血浆中盐酸二甲双胍浓度

目的建立测定人血浆中盐酸二甲双胍浓度的高效液相色谱法。方法以阿替洛尔为内标,采用Lichrospher C18色谱柱(250 mm×4.6 mm,5μm),以30 mmol/L十二烷基硫酸钠(含0.5%三乙胺,用磷酸调pH 4.0)-乙腈(65∶35)为流动相,流速为1.0 mL/min,检测波长:232 nm。结果最低检测浓度为0.010 75μg/mL,血浆中盐酸二甲双胍浓度在0.049 38~3.16μg/mL范围内与峰面积呈良好线性关系。结论此法灵敏、简便、快速、准确,适用于盐酸二甲双胍的血药浓度测定。

The determination of metformin hydochloride in human plasma by HPLC

Purpose To establish HPLC method for the determinatin of metformin hydochloride in human plasma.Methods Atenolol was used as internal standard.A Lichrospher C18(250 mm×4.6 mm,5 μm)column was and the mobile phase was composed of 30 mmol/L SDS-acetonitrile(contained 0.5% triethylamine,adjusted to pH 4.0 by phosphoric acid)(65∶35).Flow rate was 1.0 mL/min.Detection wavelength was 232 nm.Results The detection limit of metformin hydochloride was 0.010 75 μg/mL.Human plasma calibration cure for metformin hydochloride was linear in range of 0.049 38-3.16 μg/mL.Conclusion This method is sensitive,convenient,rapid and accurate.The method could be used in the determination of metformin hydochloride in human plasma and useful for pharmacokinetic study of metformin hydrochloride in clinics.
