高效液相色谱法测定血浆中盐酸二甲双胍浓度

摘    要:目的建立测定人血浆中盐酸二甲双胍浓度的高效液相色谱法。方法以阿替洛尔为内标,采用Lichrospher C18色谱柱(250 mm×4.6 mm,5μm),以30 mmol/L十二烷基硫酸钠(含0.5%三乙胺,用磷酸调pH 4.0)-乙腈(65∶35)为流动相,流速为1.0 mL/min,检测波长:232 nm。结果最低检测浓度为0.010 75μg/mL,血浆中盐酸二甲双胍浓度在0.049 38~3.16μg/mL范围内与峰面积呈良好线性关系。结论此法灵敏、简便、快速、准确,适用于盐酸二甲双胍的血药浓度测定。

Abstract:Purpose To establish HPLC method for the determinatin of metformin hydochloride in human plasma.Methods Atenolol was used as internal standard.A Lichrospher C18(250 mm×4.6 mm,5 μm)column was and the mobile phase was composed of 30 mmol/L SDS-acetonitrile(contained 0.5% triethylamine,adjusted to pH 4.0 by phosphoric acid)(65∶35).Flow rate was 1.0 mL/min.Detection wavelength was 232 nm.Results The detection limit of metformin hydochloride was 0.010 75 μg/mL.Human plasma calibration cure for metformin hydochloride was linear in range of 0.049 38-3.16 μg/mL.Conclusion This method is sensitive,convenient,rapid and accurate.The method could be used in the determination of metformin hydochloride in human plasma and useful for pharmacokinetic study of metformin hydrochloride in clinics.
