摘    要:目的 探讨ADUB EEC的部分生物学特性.方法 采用电镜、MTT、磁酶免及免疫组化等方法了解ADUB EEC的超微结构、生长曲线、CA125分泌及PCNA、ER、PR表达情况,并与在体ADUB子宫内膜组织及培养的正常EEC进行比较.结果 体外培养的ADUB EEC基本保留了体内的结构特征,体外增生水平低,分泌异常.结论 原代培养的ADUB EEC为研究无排卵功血发病机制及治疗奠定了实验基础.

Abstract:Objective To investigate the biological properties of endometrial epithelial cells(EEC) from anovulatory dysfunctional uterine bleeding(ADUB)patients. Methods EEC were isolated from 30 diagnosed ADUB patients, and from 22 patients with normal menstruation. The growth curve, ultrastructure, CA 125 production, and the expressions of proliferative cell nuclear antigen (PCNA), estrogen receptor (ER) and progesterone receptor (PR) of those cells were respectively investigated with Metromethyl Thiazolyl Blue assay (MTT), electron microscopy, MAIA and immunocytochemistry. Results ADUB EEC in vitro retained pathologic configuration of in vivo cells such as retarded growth and abnormal secretion. Conclusion The isolation and primary culture of ADUB EEC can serve to be one of the in vitro models of ADUB.
