青蒿琥酯对实验性肺纤维化大鼠治疗作用及其机理研究

摘    要:目的观察比较青篙琥酯与激素治疗肺间质纤维化的效果及作用机制。方法以博菜霉素气管内注入建立肺纤维化模型.28天造模成功后开始给予腹腔注射甲基泼尼松龙或青蒿琥酯治疗,于用药治疗后第7、14天,21、28天分批处死动物。测定血清中TGF-β1、TNF—α的浓度;通过HE染色、Masson染色判断肺组织肺泡炎及肺纤维化程度。结果与模型组相比,青蒿琥酯各剂量组、甲基泼尼松龙组的TGF-β1、TNF—α的含量显著降低(P〈0.05);但青蒿琥酯高剂量组TGF—β1、TNF—α的含量降低与甲基泼尼松龙组比较无显著差异(P〈0.05);光镜下肺组织肺泡炎、肺纤维化程度在青蒿琥酯各剂量组、甲基泼尼松龙组均比模型组减轻。结论青蒿琥酯可降低肺纤维化大鼠TGF-β1、TNF—α的表达,明显减轻肺纤维化程度,其治疗肺纤维化作用途径与甲基泼尼松龙一致。

The Study on Effect and Mechanism of Artesunate in Treating Experimental Pulmonary Fibrosis in Rats

Abstract:Objective To observe and compare the effect and the mechanism of artesunate and hormone in treating pulmonary fibrosis. Methods Pulmonary fibrosis model was constituted by injecting bleomycin in tracheas of rats, 28 days later, treated with Methyllprednisolone or artesumate injected in cavum-abdominis, killing animals on the 7^th, 14^th, 21^th, 28^th day. Determined concentration of TGF-β1, TNF-α in serum; estimated the extent of alveolar catarrh in lung tissue and pulmonary fibrosis by HE and Masson staining. Results The levels of TGF-β1 and TNF-α in dose artesunate group and Methyllprednisolone group were obviously lower than those in model group(P〈0.05); However, the levels of TGF-β1, TNF-α between high-dose artesunate group and Methyllprednisolone group had no obvious reduction (P〈0.05); The degree of alveolar catarrh in lung tissue and pulmonary fibrosis in the light microscope reduced in all dose artesunate group and methyllprednisolone group more than in model group. Conclusion Artesunate reduces expression of TGF-β1 and TNF-α in pulmonary fibrosis in rat, obviously reduces the extent of pulmonary fibrosis, and its effective ways in treating pulmonary fibrosis are consistent with methyUprednisolone.
