HPLC法同时测定无梗五加果实中金丝桃苷和东莨菪内酯的含量

目的:建立同时测定无梗五加[Acanthopanax sessiliflorus(Ruqr.et Maxim)Seem.]果实中金丝桃苷及东莨菪内酯含量的方法。方法:采用Tigerkin ODS C18,5μm,1.6×250mm色谱柱,流动相为乙腈-3%冰乙酸水溶液(体积比为20:80),流速1.0mL/min,柱温30℃,检测波长0～13min360nm,13～20min345nm。结果:金丝桃苷、东莨菪内酯的线性范围分别为0.06912～0.3456μg(R=0.9999)、0.01424～0.09968μg(R=0.9999),平均回收率分别为100.9%(RSD=2.8%)、99.6%(RSD=2.7%)。结论:此方法准确、可靠,可用于无梗五加果实的质量控制。

Determination of Hyperin and Seopoletin in the Fruit of Acanthopanax sessiliflorus(Rupr.et Maxim.)Seem.by HPLC

Objective ：To develop a HPLC method for determination of the contents of hyperin and seopoletin in fruit of A canthopanax sessiliflorus （Rupr.et Maxim.）Seem. Methods： A Tigerkin ODS C18 column （5μm,1.6×250mm） was used with a mobile phase of acetonitrile-3%glacial acetic acid gradient elution （V：V=20：80）.The flow rate was set at 1.0mL/min.The UV detection was set at 365nm at 0-13min and 345nm at 13-20min.The column temperature was 30℃.Results：The liner ranges of hyperin and seopoletin were 0.06912- 0.3456μg（r=0.9999）and 0.01424-0.09968μg（r=0.9999）,respectively. The recoveries of hyperin and seopoletin were 100.9%和99.6%. Conclusions：The method is accurate and reliable that can be used to the quality control of fruit of Acanthopanax sessiliflorus （Rupr.et Maxim.）Seem.
