桂枝茯苓丸对人宫颈癌荷瘤小鼠免疫调节机制的实验研究

摘    要:[目的]探讨桂枝茯苓丸对人宫颈癌荷瘤小鼠脾脏Th_1/Th_2细胞因子表达水平的影响。[方法]以人宫颈癌系Hela接种于BALB/c小鼠右后大腿皮下,建立宫颈癌荷瘤小鼠模型,分组后分别给予生理盐水、桂枝茯苓丸、顺氯氨铂。测定抑瘤率及小鼠胸腺指数、脾脏指数;流式细胞术检测细胞因Th_1(IL-2)/Th_2(IL-10)的表达水平。[结果]桂枝茯苓丸和顺氯氨铂对小鼠宫颈癌Hela瘤体均有明显抑制作用,能提高荷瘤小鼠脾脏指数与胸腺指数。桂枝茯苓丸治疗组,Th_1细胞因子IL-2水平显著升高,Th_1/Th_2比值明显高于模型组,而且向正常值漂移。[结论]桂枝茯苓丸有逆转宫颈癌荷瘤小鼠Th_1向Th_2漂移的作用,能恢复Th_1表达的优势,从而为桂枝茯苓丸通过免疫调节作用治疗宫颈癌提供实验依据。

Abstract:[ Objective] Study on the spleen cytokine Th1/Th2 modifying effect on human CACX tumor- bearing mice of Guizhil Fuling pill [ Methods ] We set up the tumor - bearing mice model through injecting human CACX tumor Hela ceils into right posterolateral branch of BALB/c mice, after beeing divided into groups ,we respectively give Guizhil Fuling pill and physiological saline to different groups, then add specific fluorescence labeling eytokine monoclone antibody and measure the expression of Thl (IL- 2 )/Th2( IL- 10) with the technology of flowing cytometry. [ Results ] Compared with blank group, The expression of model group mice spleen Thl cytokine IL- 2 markedly reduces ,P 〈 0.01 ,which shows that the expression of tumorbearing mice eytokine Thl attenuates ; reversely, its Th2 cytokine IL - 10 dominantly expresses, P 〈 0.01, it tells us that Th1/Th2 expression of tumor - bearing mice is maladjusted and drifts to Th2. Compared with model group, expression of Guizhil Fuling pill group mice eytokine Thl IL - 2remarkably enhances, P 〈 0.01 ; also Th1/Th2 expression is evidently above model group and drifts to normal level,whic reflects that Guizhil Fuling pill can reverse Thl drifting to Th2 on tumor - bearing mice. [ Conclusions] Guizhil Fuling pill can reverse Thl drifting to Th2 on CACX tumorbearing mice and revert the dominant expression of Th1.
