脑室注射高渗氯化钠诱导脑内c-fos与egr-1表达以及与AVP神经元的关系

摘    要:目的 观察脑室注射高渗氯化钠诱导的脑内c-fos 与egr-1 的表达特征,并用双重细胞染色方法观察脑内c-fos 表达与下丘脑视上核(SON)中加压素(AVP) 能神经元之间的关系。方法 用SD 种系清醒大鼠,侧脑室微量注射高渗氯化钠,1h 后作c-fos 与egr- 1 的免疫细胞化学染色和细胞双重染色。结果 (1) 脑室注射高渗氯化钠可在终末血管器官(OVLT)、正中视前核( MPN) 、SON、下丘脑室旁核(PVN) 以及后脑的最后区(AP) 、孤束核(STN) 和臂旁外侧核(LPBN) 中同时诱导c-fos 和egr- 1 表达,在同一区域内egr- 1 的表达略低于c-fos,但无统计学差异;(2) 穹窿下器官(SFO)中均无c-fos 和egr-1 表达;(3)SON 中可见部分AVP免疫反应阳性与c- fos表达共存于同一神经元。结论 c-fos 和egr-1 对脑内高渗刺激诱导的表达部位基本一致,进一步证实了以往的研究结果。另外,脑内注射高渗氯化钠诱导的下丘脑细胞活动至少部分是通过AVP能神经元的。

Abstract:Object The characters of expression of c-fos and egr-1 in the brain provoked by intracerebroventricular (icv) injection of hypertonic saline were observed,and the relation between Fos and AVP-containing neurons in hypothalamus was also studied by double staining.Method C-fos and egr-1 immunohistochemical and double staining were made 1h after icv injection of hypertonic saline in awaking Sprague-Dawley rats.Results Icv injection of 3 mol/L NaCl could induce Fos-like immunoreactive (FLI) and Egr-1-like immunoreactive (ELI) expression in the same areas,such as in organum vasculosum of laminae terminalis (OVLT),median preoptic nucleus (MPN) of forebrain,supraoptic nucleus (SON) and paraventricular nucleus (PVN) of hypothalamus as well as areas postrema (AP),lateral parabrachial nucleus (LPBN) and solitary tract nucleus (STN) of hindbrain.The FLI expression induced by 3 mol/L NaCl was stronger than ELI expression in the same areas,but there was no significant difference.As compared with previous studies,no ELI expression was observed in subfornic organ (SFO).Double staining showed the co-existence of c-fos and vasopressin (AVP)-cells in SON.Conclusion These results suggest that egr-1 expression induced by icv injection of hypertonic saline was in the same areas as c-fos expression in rat brain,and that the response of hypothalamic cells to icv injection of hypertonic saline is partially related to the activation of AVP-containing neurons.
