超高效液相色谱法测定钩藤中的钩藤碱和异钩藤碱的含量

摘    要:目的:用超高效液相色谱法同时测定钩藤中的钩藤碱和异钩藤碱的含量。方法:以ACQUITYUPLCR BEH130C18(2.1mm×100mm,1.7μm)为色谱柱,乙腈-0.01mol/L三乙胺水溶液(45:55,冰醋酸调pH7.0)为流动相,检测波长为245nm,流速0.2mL/min。结果:钩藤碱在0.052-0.309μg具有良好的线性关系,r=0.9997,平均回收率为98.3%,RSD为0.88%,异钩藤碱在0.060μg-0.363μg具有良好的线性关系,r=0.9999,平均回收率为97.7%,RSD为1.29%。结论:该方法简便快捷、精密度高、结果准确。可用于钩藤的质量控制。

Abstract:Objective: A method for determination of Rhynchophyline and Isorhychophylline by UPLC was created.Methods: The method contained a ACQUITYUPLCRBEH130C18 1.7μm(2.1mm×100mm Column) as column,with acetonitrile: 0.01mol/L Triethylamine solution(45:55,PH7.0 adjusted by glacial acetic acid) as mobile Phase,UV detection wavelength at 245nm.Results: The calibration curve of Rhynchophyline was linear between 0.052-0.309μg,r=0.9997,the average recovery was 98.3%,RSD=0.88%.The calibration curve of Isorhychophylline was linear between 0.060μg-0.363μg,r=0.9999,the average recovery was 97.7%,RSD=1.29%.Conclusion: This method was proved to be simple,rapid,accurated and can be used for the quality control of Uncaria rhynchophylla.
