益髓颗粒对免疫性血小板减少性紫癜模型小鼠细胞相关因子表达影响

摘    要:目的:建立免疫性血小板减少性紫癜(ITP)小鼠动物模型,观察益髓颗粒对模型小鼠外周血细胞因子影响,探讨益髓颗粒的免疫调节机制。方法:40只Balb/c小鼠随机分为正常组、模型组、泼尼松治疗组和益髓颗粒治疗组,除正常组外,其余各组隔日1次腹腔注射豚鼠抗小鼠血小板血清(GP-APS)建立ITP小鼠模型,于造模第8d开始,各组均按0.2mL/(10g·d)体积灌胃。其中正常组、模型组灌服生理盐水;泼尼松按0.0113mg/(10g·d)灌胃;益髓颗粒按0.3g/(10g·d)灌胃。连续7d后眼球取血分离血清,酶联免疫吸附法(ELISA)检测干扰素-γ(IFN-γ)、白细胞介素-4(IL-4)。结果:与正常组相比,模型组小鼠IFN-γ水平明显升高,IL-4水平明显降低,IFN-γ/IL-4比值明显异常;与模型组相比,泼尼松治疗、益髓颗粒治疗组的IFN-γ、IL-4均有所恢复,IFN-γ/IL-4比值接近正常。结论:益髓颗粒可通过影响Th1/Th2细胞及其因子水平而对ITP免疫功能起调节作用。
