摘    要:应用荧光探剂ANS,研究了氯化汞与大鼠脑突触体膜的相互作用。结果发现,氯化汞增强膜ANS复合物荧光强度。双倒数及Scatchard作图分析表明,其使膜-ANS复合物荧光强度增强的原因并非ANS荧光量子产率的改变,而是增加了突触体膜对ANS的结合容量和亲和力,亦即降低了膜表面负电荷密度。这一效应与其对膜流动性的影响是一致的。

Abstract:Interactions of mercuric chloride with rat brain synaptosomal membranes were studied with 8-anilino-1-naphthalene sulphonic acid (ANS)as a fluorescent probe. It was found that mercuric chloride enhanced the fluorescence intensity of the ANS-membrane complex. A graphical analysis of this effect shows it to be due to an increase in the amount of ANS bound to the membrane, not to an increase in the quantum yield of ANS. The Scatchard plots further confirms that mercuric chloride increases the number of ANS binding sites and affinity of ANS for the membranes,namely decreases the negative charge of the membrane surface. This effect is consistent with its effect on the lipid fluidity of synapto-somes.
