xylE转基因小鼠的繁育及其纯合子的筛选

摘    要:目的 进行转基因小鼠的繁育与纯合子转基因小鼠的筛选。方法 选用致突变xylE转基因原代小鼠31号和9号进行传代,自F2起采用全同胞交配繁育至F3。结果 转基因能遗传且各世代转基因小鼠表型无异常,亦未见近交衰退现象。经不同世代载体回收和转化检验表明,靶基因功能未因世代传递而改变,并从3l号系的F3中筛选出一只纯合雄鼠,经与非转基因雌鼠交配所产4窝28只小鼠,PCR—southern检测均呈阳性,证实为~纯合子。结论 xylE转基因能遗传,4个世代内便筛选出纯合子小鼠,极大地缩短了育种周期。

Abstract:Objective To breed transgenic mice containing xylE gene and to screen homozygous transgenic mice. Methods Two founders(G 09 and G 031) of transgenic mice containing a %xylE% mutational target gene were selected for breeding. G 09 and G 031 were mated with nontransgenic ICR mice respectively and F 1 offsprings were produced. F 2 and F 3 offsprings were generated by inbreeding of the same litters of F 1 or F 2 respectively. Results Their offspring's phenotype had no detectable abnormal. Detection of line 31 indicated that the %xylE% target gene could stably transmit to the next generations. One homozygous male mouse of line 31 by PCR had been confirmed. Through mating the mouce to nontransgenic partners, all the 28 offsprings (four litters) were transgenic by PCR-Southern detection. Conclusions 1.Transgene could be stably transmitted to F 3. 2.One homozygous transgenic mouse was screened within 4 generations of line 31.;
