应用体外抑菌法筛选中华大蟾蜍抗结核有效成分的研究

摘    要:目的验证中华大蟾蜍具有抗结核活性,筛选抗结核活性成分。方法对中华大蟾蜍水提物进行葡聚糖凝胶分子筛分离和反相高效液相层析法(RPLC)分离,并采用微孔板Alamarblue法将分离所得12组分进行体外抑菌实验研究。结果第1、6、10组分表现出对供试结核分枝杆菌的明显抑制作用,其最低抑菌浓度(minimal inhibitory concentration,MIC)分别为339.98mg/ml、20.00mg/ml、2.95mg/ml。结论中华大蟾蜍含有抑制结核分枝杆菌的活性成分,该有效成分分子量接近14KD,并表现出弱极性的特征。

Abstract:Objective To validate the potential antimicrobial activity of Bufo bufo gargarizans cantor against tuberculosis and to screen its active components. Methods Bioassay-guided fractions were done with the method of gel filtration chromatography and RPLC.
