联合应用hIL-2和hGM-CSF基因瘤苗抗肿瘤作用的研究

摘    要:目的:探讨联合应用人白细胞介素2(hIL-2)和人粒细胞巨呼细胞刺激因子(hGM-CSF)基因瘤苗抗肿瘤作用。方法:经逆转录病毒载体将hIL-2基因及hGM-CSF基因导入大鼠Walker瘤细胞WRC256中,制备瘤苗接种荷瘤大鼠。结果:经PCR-Southerm Blot检测证实hIL-2基因及hGM-CSF基因已成功导入WRC256细胞基因组中,经生物学活性检测证实转基因细胞具有分泌hIL-2及hGM-CSF活性。转基因瘤苗经60Gy X线灭活,接种荷瘤Walker瘤模型,两种瘤苗确可诱导有效抗肿瘤反应,但与单独应用一种瘤苗相比较,联合应用无相加或协同作用。结论:两种瘤苗可诱导有效抗肿瘤反应,但联合应用无相加或协同作用。

Abstract:Objective: To investigate the Anti tumour effect of the tumour vaccines engineered by human interleukin 2(hIL 2) and human granulocyte macrophage colony stimulating factor(hGM CSF) genes. Methods: The hIL 2 and hGM CSF genes were transferred into the Walker tumour cell vaccinesbby virtue of retrovirus. The gene transferred vaccines were inoculated into the Walker tumour model.Results:Gene transfers were successfully confirmed by PCR Southern Blot. The biological activities of hIL 2 and hGM CSF by the gene transferred vaccine cells were showed with MTT assay. After in Vitro irradiation with 60Gy X ray, the gene transferred vaccines were inoculated into the Walker tumour model. Conclusion:The effective anti tumour response was induced by the vaccines. The plus or coordinate anti tumour effect by synchronous inoculation with two kinds of the vaccines was not observed.
