Abstract:This study was designed to investigate the effect of IL-35 on T cell phenotype and cancer progression in lung cancer.The tumor model of lung cancer was established in mice,and the tumor growth was monitored after administration of IL-35 neutralizing antibody.The anti-tumor immune response was further explored by detecting the infiltration of T cells and cytokines production in the tumor.Finally,the anti-tumor activity of infiltrating T cells was detected by activation experiment in vitro.The detection results of tumor volume and tumor weight showed that IL-35 could inhibit the growth of lung tumor(P<0.05).At the same time,neutralizing IL-35 could also lead to less regulatory T cell infiltration(P<0.01),an increase in the proportion of cytotoxic T cells,decrease in the expression of inhibitory receptors on T cells(P<0.001)and increase in the production of IFN-γand TNF-α(P<0.01).In addition,the in vitro activation ability of tumor infiltrating T cells neutralized by IL-35 was also significantly enhanced(P<0.001).In conclusion,the neutralization of IL-35 can inhibit the growth of lung tumor,which is partly due to the decrease of tumor infiltrating regulatory T cells and the reversal of T cell exhaustion,thus enhancing the anti-tumor immune response.
