参芪化瘀方对H22肝癌细胞p53基因表达的影响

摘    要:目的:观察参芪化瘀方对荷瘤小鼠肿瘤组织p53基因表达的影响,探讨参芪化瘀方对H22肝癌细胞作用的机制。方法:将H22肝癌荷瘤小鼠60只随机分为6组各10只:参芪化瘀方高剂量组、中剂量组、低剂量组,环磷酰胺注射用粉针剂(CTX)对照组,模型组,阴性对照组。参芪化瘀方高、中、低剂量组予以0.4mL中药(浓度分别为4.16g/mL、2.08g/mL、1.04g/mL)灌胃,CTX对照组予以0.2mL[小鼠给药剂量为20mg/(kg·d)]腹腔注射,模型组予以0.4mL生理盐水灌胃,每天1次,给药14天。免疫组织化学法检测肿瘤组织p53基因的表达,并观察其对脾脏指数、胸腺指数及肿瘤抑制率。结果:高、中、低剂量参芪化瘀方都能降低H22肝癌荷瘤小鼠肿瘤组织中p53基因的表达水平,且能升高小鼠的脾脏指数、胸腺指数。结论:参芪化瘀方对H22肝癌荷瘤小鼠肿瘤组织中p53基因的表达具有一定抑制作用,且能保护小鼠的免疫器官。

Abstract:Objective. To observe the effect of Shenqi Huayu Formula on p53 gene expression in tumor-bearing mice, and to explore its mechanism in influencing H22 hepatoma cells. Methods: Sixty mice transplanted with H22 hepatoma cells Were randomly divided into six groups: high-, middle- and low-dose Shenqi Huayu Formula groups (gastric gavage of 0.4mL of the decoction of Shenqi Huayu Formula), cycloohosphamide (CTX) group [intraperitoneal injection of 0.2mL of CTX at the concentration of 20mg/(kg ·d)], model group (gastric gavage of 0.4mL of normal saline), and negative control group. The medication was carried out once a day and lasted 14 days. The expression of p53 gene in the tumor tissue was tested by immunohistochemical assay. Spleen index, thymus index and tumor-inhibition rate were also observed. Results: The expression of p53 gene in all of Shenqi Huayu Formula groups was reduced, and the spleen index and thymus index were increased. Conclusion. Shenqi Huayu Formula can inhibit the expression of p53 gene and protect the immune organs of H22 tumor-bearing mice.
