星点设计-效应面法优化解毒通络颗粒的提取工艺

摘    要:目的:优选解毒通络颗粒的提取工艺。方法:选择浸泡时间、煎煮时间和溶剂用量为自变量,黄芩苷、槲皮素、异鼠李素提取量的综合评分为因变量,在单因素试验基础上,采用星点设计-效应面法优选提取工艺,通过SPSS软件对试验数据进行多元线性模型和二次多项式模型拟合,确定最佳数学模型,利用Design Expert 8.0.0统计分析软件绘制效应面图和等高线图。结果:最佳提取工艺为浸泡时间90 min,煎煮时间85.63 min,溶剂用量9.21倍;综合评分(93.2±0.051)%,与预测值(94.30%)的偏差仅-2.32%。结论:通过星点设计-效应面法建立的模型预测性良好,适用于解毒通络颗粒提取工艺的优选。

Abstract:Objective: To optimize extraction technology of Jiedu Tongluo particles. Method: Based on single factor tests, central composite design-response surface methodology was applied to optimize extraction process with soaking time, decoction time and solvent amounts as independent variables, while composite score of extraction amounts of baicalin, quercetin and isorhamnetin as dependent variable.SPSS software was used to fit multivariate linear equation and second-order polynomial equation for experimental data, response surface and contour plot were delineated according to best-fit mathematic models by Design Expert 8.0.0 statistic analysis software. Result: Optimum extraction process was as follows:soaking time 90 min, decoction time 85.63 min, the amount of solvent 9.21 times;Composite score was (93.2±0.051) %, which had deviation of -2.32% with the predicted value of 94.30%. Conclusion: Model established by central composite design-response surface methodology had good predictability, which was suitable for optimizing extraction process of Jiedu Tongluo particles.
