诃子提取物对过氧化氢所致乳鼠心肌细胞损伤的保护作用

摘    要:目的研究诃子提取物对过氧化氢(H2O2)所致心肌细胞损伤的保护作用。方法采用H2O2处理体外培养心肌细胞,造成氧化应激模型,通过检测细胞培养液中乳酸脱氢酶(LDH)、肌酸激酶(CK)漏出量,丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性来反映诃子提取物对H2O2氧化模型的影响,并采用四甲基偶氮唑盐比色法(MTT法)测定细胞的代谢率。结果与模型组相比,诃子提取物能显著减少LDH和CK漏出量(P〈0.01),升高A值(P〈0.01),降低培养液中MDA含量(P〈0.05,P〈0.01),升高SOD活性(P〈0.01)。结论诃子提取物可减轻H2O2对心肌细胞的损伤程度,对心肌细胞具有保护作用,其机制可能与稳定细胞膜和抗氧化有关。

Abstract:Objective To investigate the protective effects of the Terminalia chebula extract on cultured rat cardiocyte injury induced by hy-drogen peroxide(H2O2).Methods The oxidative stress model was established by treating the cultured rat myocardial cells in vitro with H2O2.The leakage level of lactic dehydrogenase(LDH),creatine kinase(CK),the content of malondialdehyde(MDA) and the activity of su-peroxide dismutase(SOD) in cell supernatant were measured to observe the effects of the Terminalia chebula extract on the H2O2-induced oxidative model.MTT was adopted to detect the cell metabolic rate.Results Compared with the model control,the Terminalia chebula extract significantly decreased the leakage level of LDH and CK(P 0.01),increased A value significantly(P 0.01),decreased the content of MDA in cell supernatant significantly(P 0.05,P 0.01) and increased the SOD activity significantly(P 0.01).Conclusion the Termi-nalia chebula extract can mitigate the injury of H2O2 on cardiocytes with the protecting effect on damaged myocardium,which mechanism may be related to stabilize membrane and its antioxidant properties.
