斯氏狸殖和丰宫并殖吸虫成虫乳酸脱氢酶同工酶等电聚焦电泳的比较研究

摘    要:目的通过对云南省斯氏狸殖吸虫(Pagomogonimus skrjabini Chen.1959)和丰宫并殖吸虫(Paragonimus proliferus.1964)成虫乳酸脱氢酶(LDH)的比较研究,为吸虫的分类及定种提供科学依据方法采用丙烯酰胺凝胶圆盘等电聚焦电泳,对乳酸脱氨酶同工酶进行分离,并采用底物特一性染色法进行确定电泳结果。结果两虫种成虫乳酸脱氢酶同工酶谱分析,结果发现二者均显示5条酶带,其中斯氏狸殖吸虫的LDH有3条优势酶带(LDD_1、LDH_2、LDH_3)而丰宫并殖吸虫的LDH仅有2条优势酶带(LDH_1、LDH_2);对各成分的等电点及相对迂移率进行计算,发现丰宫并殖吸虫与斯氏狸殖吸虫间存在生化指标的差异,同时也发现两虫种间具有一共同性质的LDH_4。结论通过对两虫种同工酶谱的分析可知,它们之间具有一定的亲源性,但二者间又有同工酶谱的差异性,因此乳酸脱氢酶同工酶谱的分析可为并殖吸虫的分类提供生化依据和指标。

Abstract:Objective comparative study on Isoenzyme of Lactate Dehydrogenase (LDH) of Pagomogonimus skrjabini chen.(1959) Adults And Paragonimus proliferus(1964) Adults for offering scientific characteristics of classification. Methods separate Isoenzyme of Lactate Dehydrogenase (LDH)of Pagomogonimus skrjabini chen.(1959) Adults And Paragonimus proliferus (1964) Adults, which collected from Jinghong county and Jinping county, Yunnan, by polyacrylamide gel disc-Isoelectrofocusing(IEF),. and analyze the results by Specific Substrate Reaction dyeing. Results In this study we found there were 5 bands of isoenzyme of LDH in two species homogenized samples. Three prevailing bands (LDH1, LDH2, LDH3) found in homogenized samples of Pagomogonimus skrjabini chen., two prevailing bands (LDH1, LDH2) found in homogenized samples of Paragonimus proliferus. We found one component, LDH4, possessed the same PI, Analyzing relative mobility, fraction of the eletrophoresis result, we observed the differences between two species. Conclusions Through this study we could learn that there were some common characteristics between Pagomogonimus Skrjabini chen. and Paragonimus proliferus, as well as there were some differences between them. We can use these biochemical characteristics of LDH in Paragonimus clssification.
