黑米花色苷提取物在小鼠哮喘模型中对p38 丝裂原活化蛋白激酶信号通路的影响

摘    要:目的 探讨黑米花色苷(AEBR)提取物对哮喘小鼠气道炎症和 Thl/Th2 类细胞因子变化的影响及其可能机制.方法 雄性 BALB/c 小鼠32只随机分成4组,即正常对照组、哮喘模型组和黑米花色苷提取物低、高剂量干预组.分别采用酶联免疫吸附法 (ELISA) 和免疫印迹法检测支气管肺泡灌洗液 (BALF) 中白细胞介素-4、白细胞介素-5、白细胞介素-13(IL-4、IL-5、IL-13) 及干扰素-γ(IFN-γ) 含量以及肺组织中磷酸化p38丝裂原活化蛋白激酶(p38 MAPK)的变化,并观察 BALF 中炎症细胞和肺组织病理学改变.结果 哮喘模型组小鼠支气管肺泡灌洗液中炎症细胞计数、白细胞介素-4、白细胞介素-5、白细胞介素-13水平升高和干扰素-γ水平降低;肺组织中磷酸化p38丝裂原活化蛋白激酶表达水平升高,与正常对照组比较,差异有显著性(P<0.05).黑米花色苷提取物低、高剂量干预组小鼠BALF中炎症细胞计数、白细胞介素-4、白细胞介素-5、白细胞介素-13 水平和肺组织中磷酸化 p38丝裂原活化蛋白激酶表达水平均明显降低,IFN-γ水平明显上升,与哮喘模型组比较,差异均具有显著性 (P<0.05).黑米花色苷提取物处理能明显减轻哮喘小鼠肺组织病理学改变,黑米花色苷提取物低、高剂量干预组之间比较,差异不显著 (P>0.05).结论 p38丝裂原活化蛋白激酶可能参与了支气管哮喘的发病过程.黑米花色苷提取物对哮喘小鼠具有保护作用,其机制可能与抑制 p38丝裂原活化蛋白激酶磷酸化、调节白细胞介素-4 / 干扰素-γ平衡失调、减轻炎症细胞浸润有关.

Abstract:Objective To explore the protective effects and mechanism of anthocyanin-rich extract from black rice(AEBR)in asthmatic mice. Methods Ovalbumin(OVA) was injected intraperitoneally and inhaled to produce the asthmatic model. Thirty two mice were randomly divided into four groups:control group, asthma group and AEBR groups of high and low dose. The concentrations of IL-4、IL-5、IL-13 and IFN-γ in bronchoalveolar lavage fluid(BALF) were respectively measured by ELISA. The phosphor-p38 mitogen-activated protein kinases(MAPK) in lung tissues was measured by Western blotting. The number of inflammatory cells in BALF and lung histropathological changes were observed Results In asthmatic group,the number of inflammatory cells and the concentrations of IL-4、IL-5、IL-13 in BALF and phosphor-p38 MAPK in lung tissue were higher, but IFN-γ was lower than those in control mouse (EM>P/EM> <0.05). In AEBR group, the number of inflammatory cells, the concentrations of IL-4、IL-5、IL-13 in BALF and phosphor-p38 MAPK in lung tissues were higher, but IFN-γ was lower than those in normal control mice(EM>P/EM> <0.05), and histropathology damage was alleviated significantly.
