摘    要:目的 检测胆道闭锁(biliary atresia,BA)患儿肝脏组织中Toll样受体(TLRs)的表达情况,探讨其与胆道闭锁发生的关系.方法 应用RT-PCR和Westen blot方法检测13例BA患儿、7例胆总管扩张(CC)患儿和8例正常对照儿童(CO)肝脏组织中TLRs mRNA和蛋白的表达.结果 BA患儿肝脏组织TLR7表达明显高于CC组患儿和CO组患儿(P<0.05);而TLR3的表达在BA组患儿和CC组患儿之间差异无统计学意义,略高于CO组患儿(P>0.05).TLR4的表达在BA组患儿和CC组患儿之间差异无统计学意义(P>0.05),但高于CO组患儿(P<0.05).结论 胆道闭锁患儿肝组织中TLR3、TLR7、TLR4表达异常,可能在胆道闭锁胆管损伤中发挥作用.

Abstract:Objective To explore the expression of Toll-like receptors in 1ivers of patients with biliary atresia(BA)and tO explore the relationship between Toll-like receptors and BA.Methods Expression of mRNA and protein of Toll-like receptors in livers of 13 patients with BA.7 with choledochal cyst(CC)and eight normal control(CO)children was detected by means of RT-PCR and western blot.Results The expression of TLR7 was significantly higher in patients with BA than in those with CC and CO(P<0.05);TLR3 expression was a little higher in patients with BA than in CO(P>0.05),and no statistically significant difference could be noted between patients with BA and CC(P>0.05):there was no statistically significant difference of the expression of TLR4 between patients with BA and CC(P>0.05),while the expression of TLR4 was significantly higher in patients with BA than in CO(P<0.05).Conclusions There iS abnormal expression of Toll-like receptors in the livers of patients with BA.A possible explanation iS that TLR3,TLR7,TLR4 may play important roles in the lesion of biliary tract in patients with BA.
