摘    要:合成一对恶性疟原虫特异引物,用聚合酶链的反应(PCR)技术检测海南岛恶性疟病人滤纸血滴30例,结果28例阳性,在206bp处可见一条清晰易辨的扩增条带,2例阴性经血片复查,其中1例为间日疟原虫;同时检测间日疟病人血滴和未发现原虫的发热病人血滴各16例,均为阴性,与镜检符合率达98.4%(61/62例)。初步表明PCR法具有准确、敏感、特异、简捷等优点,采用滤纸血滴方便现场采样、保存和送检,且易批量检测,在疟疾临床诊断和流行病学调查方面,将可替代血片镜检法,逐步扩大应用。

Abstract:Sixty-two filter paper blood samples from patienls with malaria or fever were detected by polymerase chain reaction (PCR) using a pair primers of which amplify a 206 bp sequence from repetitive genomic DNA of Plasmodium falciparum. The results show that 28 samples from patients with Plasmodium faleiparum malaria were positive, two samples were negative (blood examination showed that one sample was from a patient infected by Plasmodium vivax) while both 16 blood samples from patients with Plasmdium vivax malaria and 16 ones from smear-negative feverish patients were negative. The concordance of PCR with conventional microscopy was 98.4%.This results suggest that PCR is an accurate, specifie, sensitive and simple method.Again it is easy to collect, preserve and transport the blood samples and detect in batches. In addition, it could be used in epidemiologic investigation and clinical diagnosis of malaria in place of microscopy.
