咪达唑仑对东莨菪碱致大鼠认知功能障碍的影响

摘    要:目的 观察咪达唑仑对东莨菪碱致大鼠认知功能障碍的影响。方法 健康成年雄性sD大鼠74只,体重150—170g,随机分为4组:对照组(Con组,n=18)、咪达唑仑组(Mid组,n=18)、东莨菪碱组(Sco组,n=19)、咪达唑仑+东莨菪碱组(Mid+SCO组,n=19)。Mid组、Mid+sco组大鼠每天腹腔注射咪达唑仑50mg/kg,连续3d,记录两组翻正反射消失时间及翻正反射恢复时间。每组的一半大鼠在末次注射咪达唑仑后1d开始进行Morris水迷宫测试,另外的一半大鼠在末次注射咪达唑仑后7d开始进行Morris水迷宫测试,连续3d,Mid+SCO组、Sco组大鼠在每天水迷宫测试前20min腹腔注射东莨菪碱0.8mg/kg。结果 Mid+sco组和Mid组每天注射咪达唑仑后翻正反射消失时间及翻正反射恢复时间差异无统计学意义;与Con组及Mid组比较,Sco组大鼠出现了认知功能障碍;与Sco组比较,Mid+sco组末次注射咪达唑仑后2、3d认知功能障碍加重,而末次注射咪达唑仑后7~9d认知功能测试结果差异无统计学意义。结论 腹腔注射咪达唑仑50mg/kg(连续3d)可短暂加重东莨菪碱致大鼠认知功能障碍。

Abstract:Objective To investigate the effect of midazolam on cognitive dysfunction induced by scopolamine in rats.Methods Seventy-four male SD rats weighing 150-170 g were randomly divided into 4 groups:group A control(n=18);group B midazolam(n=18);group C scopolamine(n=19)and group D midazolam+scopolamine(n=19).In group B and D midazolzm 50 mg/kg was injected intraperitoneally once a day for 3 consecutive days.In group C and D scopolamine 0.8 mg/kg was injected intraperitoneally 20 min before Morris water-maze test(MWM test).Half of the animals in each group underwent MWM test once a day for 3 consecutive days starting from the 1st day and the other half starting from the 7 th day after the last IP midazolam. Results There was no significant difference in the duration of loss and recovery of righting reflex between group B (midazolam)and D(midazolam+scopolamine).Cognitive dysfunction was induced by scopolamine in group C as compared with group A.It was demonstrated by MWM test performed on the first 3 days after last midazolam that cognitive dysfunction induced by scopolamine was aggravated by midazolam in group D as compared with group C. Conclusion Midazolam can aggravate transiently the cognitive dysfunction induced by scopolamine in rats.
