艾叶油对小鼠胚胎骨骼发育的影响

摘    要:目的探讨艾叶油是否对小鼠胚胎骨骼发育有毒性作用。方法选用昆明种孕鼠25只,随机分为5组:正常对照组,ig给予等容积花生油;艾叶油2,1和0.5ml·kg-1组,ig给予艾叶油;环磷酰胺12.5mg·kg-1阳性对照组。艾叶油组和正常对照组自孕第12天开始ig给药,连续5d;阳性对照组只于孕第13天ip给予环磷酰胺。各组孕鼠均于孕第16天处死后取出胎鼠测量身长和尾长,将1/2胎鼠行阿利新蓝和茜素红骨骼双染,对前肢芽进行Neubert评分,观察胎鼠主要骨骼骨化点的发育。结果艾叶油2,1和0.5ml·kg-1组胎鼠身长分别为18.9±0.4,19.4±1.4,(19.9±1.0)mm,尾长分别为7.8±0.2,8.5±0.7,(8.5±0.7)mm,与正常对照组身长(19.0±1.0)mm和尾长(7.9±1.1)mm相比无显著性差异;艾叶油2,1和0.5ml·kg-1组前肢芽Neubert评分分别为285.0±3.5,278.0±4.1和289.4±2.6,与正常对照组294.0±0.8相比无显著性差异;与正常对照组相比,艾叶油2,1和0.5ml·kg-1组主要骨骼发育骨化点出现的数量无显著性差异;而阳性对照环磷酰胺组胎鼠身长和尾长明显缩短(P<0.05),分别缩短了21%和23%,前肢芽Neubert评分及骨骼发育骨化点出现的数量也显著低于艾叶油组(P<0.05)。结论艾叶油2,1和0.5ml·kg-1对胎鼠肢芽和骨骼发育无毒性作用。

Abstract:OBJECTIVE To explore whether volatile oil from Artemisia argyi has toxicity on skeleton of embryo mice. METHODS Twenty-five Kunming pregnant mice were randomly divided into 5 groups: normal control group, ig given with peanut oil in equal volume; volatile oil 2, 1and 0.5 ml·kg-1 groups, ig given volatile oil; and cyclophosphamide (positive control) group, ip given cyclophosphamide 12.5 mg·kg-1. The volatile group and normal control group were ig administrated once a day from the 12th day of pregnancy to the 16th day, for 5 d, while positive control group was ip given cyclophosphomide on the 13th day of pregnancy for 1 d. On the 16th day, the pregnant mice in each group were sacrificed to take out the embryo mice, and their body length and cauda length were measured, and half of the fetuses were observed for main skeleton development by double staining with alcian blue and alizari red S, scoring the forelimb buds with Neubert method. RESULTS The body length was 18.9±0.4, 19.4±1.4 and (19.9±1.0)mm, respectively, and th cauda length was 7.8±0.2, 8.5±0.7 and (8.5±0.6)mm, respectively in volatile oil groups, compared with was body length (19.0±1.0)mm and caude length (7.9±1.1)mm in normal control group, there was no significant difference. There was also no difference in the Neubert score of the forelimb buds between in volatile oil groups(285.0±3.5,278.0±4.1 and 289.4±2.6, respectively)and in normal control group(294.0±0.8). Also, compared with normal control group, main skeleton ossification of fetuses had no significant difference in volatile oil groups. Compared with normal control group and volatile oil groups, the body length and the cauda length were shortened dramatically by 21% and 23% in cyclophosphamide group. Compared with normal control group and volatile oil groups, the Neubert score of the forelimb buds and skeleton ossification of fetuses in cyclophosphamide group also showed significant difference(P<0.05). CONCLUSION Volatile oil from A. argyi 2, 1 and 0.5 ml·kg-1 have no toxicity on forelimb buds and skeleton of embryo mice.
