摘    要:目的在建立中度颅脑损伤动物模型的基础上,通过观察创伤后高压氧治疗对大鼠基因mdm2、bcl-2和bax表达的影响,探讨高压氧治疗作用的分子机制,为高压氧在临床上的应用提供理论依据。方法成年健康雌性SD大鼠16只,随机分为两组(脑损伤组,高压氧治疗组)。各组大鼠均于致伤后48h,取挫伤灶周边脑组织,采用免疫组化法测定基因mdm2、bcl-2和bax的表达。结果高压氧治疗组基因bcl-2的表达强于损伤组(P〈0.05)。而基因mdm2、bax在两组间的表达无差异。结论高压氧对脑损伤的治疗作用与抑制细胞凋亡有密切关系。

Effects of hyperbaric oxygen an expression of mdm2, bcl- 2 and bax after traumatic brain injury

Abstract:Objective To explore the molecular mechanism of hyperbaric oxygen (HBO) treatment of moderate craniocerebral injury in animal models and the influence of HBO on expression of mdm2, bcl - 2 and bax after traumatic brain injury (TBI). Methods 16 healthy female SD rats were randomly allocated into two groups (TBI group and HBO group). The brain tissues of rats obtained at 48 hours after cerebral traumatic operation were examined by immunohistochemical method for the expression of mdm2, bcl - 2 and bax, Results The expression of bcl - 2 in rats of HBO group was stronger than those of TBI group( P 〈 0.05 ), There was no difference in expression of bax and mdm2 in rats of these two groups. Conclusion The therapeutic effect of HBO on TBI is closely related to the suppression of apoptosis of neurons.
