摘    要:目的探讨肿瘤细胞RNA和趋化因子CXCL10基因共同修饰的树突状细胞(DC)瘤苗对小鼠抗肿瘤免疫反应的诱导作用。方法将小鼠RM 1前列腺癌细胞RNA加载小鼠骨髓来源的DC后,通过脂质体转染法将CXCL10基因导入DC,制备前列腺癌瘤苗;体内外实验检测该瘤苗诱导的抗前列腺癌免疫治疗作用。结果转染CXCL10基因的DC上清对淋巴细胞有较强的趋化作用;DC瘤苗所诱导的特异性效应细胞(CTL)对RM 1细胞具有特异性杀伤作用;经DC瘤苗治疗的荷瘤小鼠模型瘤体生长减慢,存活期延长;该瘤苗还具有明显的免疫保护作用。结论构建的前列腺癌DC瘤苗能够有效诱导抗肿瘤免疫反应和免疫保护功能。
