大鼠内脏脂肪含量与ghrelin分泌的关系

目的了解大鼠内脏脂肪含量对ghrelin分泌的影响。方法80只大鼠分别制成正常大鼠模型组、高脂大鼠模型组及减重大鼠模型,使用GE Lightspeed 16排螺旋CT全扫,计算大鼠内脏脂肪组织面积,从而计算出内脏脂肪含量,并根据内脏脂肪含量将所有大鼠重新分为A组(内脏脂肪含量<3%)、B组(3%≤内脏脂肪含量<6%)、C组(6%≤内脏脂肪含量<9%)、D组(内脏脂肪含量≥9%),应用放射免疫测定法测定血清中ghrelin的含量,进一步分析大鼠内脏脂肪含量与ghrelin分泌之间的关系。结果(1)与正常饲料喂养大鼠相比,高脂饮食后大鼠体质量与内脏脂肪含量均明显增加(P<0.01);而肥胖大鼠减重后,体质量与内脏脂肪含量均下降(P<0.01),尤其内脏脂肪含量下降更为明显(P<0.01)。(2)与A组内脏脂肪含量相比,B组、C组、D组内脏脂肪含量分别是A组的3.15倍、5.84倍、7.69倍。B组与A组之间ghrelin的分泌无显著差异(P>0.05);C组、D组ghrelin的分泌较A组明显下降(P<0.01)。结论不同内脏脂肪含量的大鼠,其ghrelin的分泌不同,在内脏脂肪含量相差较明显时,ghrelin分泌的下降才具有显著差异。

Relationship between visceral adipose and secretion of ghrelin in rats

Objective To study the effect of visceral adipose on secretion of ghrelin.Methods Eighty rats with different visceral adiposity were obtained through high fat feeding and weight loss method and were divided into normal model group,hypso-fat model group and weight relief group.All rats underwent GE Lightspeed helical CT.The area of visceral adipose tissue were calculated on helical CT picture and worked out the content of visceral adipose.All rats were divided into four groups according the content of visceral adiposity(VAT):group A(VAT<3%),group B(3%≤VAT <6%),group C(6%≤VAT <9%)and group D(VAT ≥9%).Radioimmunoassay was used to measure the plasma level of ghrelin,and the effect of visceral adiposity on the secretion of ghrelin was further to be analyzed.Results(1)Compared with the rats which obtained normal feeding,the visceral adiposity in the rats obtained high fat feeding was significantly higher(P<0.01).The weight and visceral adipose all decreased after the obese rats obtained weight loss method(P<0.01),and the visceral adiposity decreased obviously(P<0.01).(2)Compared with group A,the visceral adiposity in group B,group C and group D were 3.15,5.84 and 7.69 times more than that in group A respectively.The secretion of ghrelin was no difference between group B and group A(P>0.05).Compared with group A,the secretion of ghrelin in group C and group D decreased obviously(P<0.01).Conclusion The secretion of ghrelin is different when the rats obtain different visceral adipose.The secretion of ghrelin decrease significantly only when the contrast of visceral adiposity is significntly.
