超声定位辐照载药微泡治疗卵巢癌移植瘤的实验研究

摘    要:目的观察超声定位辐照载紫杉醇脂质微泡(paclitaxel—carrying liposome microbubbles,PLM)介导药物释放的方法对裸鼠人卵巢癌移植瘤的抑制效应。方法对荷瘤鼠进行分组处理,包括超声定位辐照PLM组、单纯静脉给药组、超声定位辐照脂质微泡组、单纯静脉输注PLM组和生理盐水对照组。处理结束后剖取瘤块测其体积、质量,计算抑瘤率;并用免疫组化法检测肿瘤组织血管内皮生长因子(VEGF)表达。结果各组的肿瘤体积、抑瘤率及VEGF表达均有差异(P<0.01)。超声定位辐照PLM组的肿瘤体积、质量最小,抑瘤率最大,VEGF表达量最低。结论超声定位辐照PLM的方法能有效介导药物释放,具有较强的体内抑瘤效果,有望成为新的肿瘤化疗给药方式。

Abstract:Objective To explore the anti-tumor effects of paclitaxel-carrying microbubbles(PLM) on ovarian cancer-bearing nude mice using the technique of ultrasound mediated drug release.Methods Ultrasound irradiation was applied to the tumors after injecting PLM through the tail vein of nude mice.After seven-day treatment,all the mice were sacrificed and the tumors were harvested to measure volumes and masses.The tumor inhibition rate was calculated by mass and the expressions of vascular endothelial growth factor (VEGF) inside the tumor tissues were detected by immunochemical staining.Results The mean tumor volume and mass in the ultrasound combined PLM group was the lowest among all the groups(4 tested groups and the control group,5 mice in each group),while its tumor inhibition rate was the highest(P<0.05) and the mean VEGF expression in this group was the lowest (P<0.05).Conclusions Paclitaxel was efficiently released from PLM mediated by ultrasound.This technique enhanced the drug delivery efficiency at the tumor site,which inhibitted the tumor growth significantly in vivo.This technique is possible to be adopted as a novel tool for ovarian cancer chemotherapy.
