血脉通胶囊对急性心肌梗死大鼠心肌病理形态学的影响

摘    要:目的: 研究血脉通胶囊(XMT)对大鼠急性心肌梗死(AMI)心肌病理形态学的影响。方法: 大鼠随机分为模型组、假手术组、血脉通胶囊低、中、高剂量组(105,210,420 mg·kg-1)。血脉通胶囊各给药组连续灌胃给药14 d后,除假手术组外,其余各组均结扎大鼠冠状动脉建立AMI模型。记录结扎前及结扎后15, 30, 60, 90, 120 min,24 h心电图变化;HE染色观察心肌病理形态学改变。结果: 和模型组比较,血脉通胶囊各剂量组可不同程度对抗AMI心电图J点的上抬;模型组大鼠心肌内膜局部排列紊乱,肌细胞显肿胀、坏死,外膜灶状坏死,间质水肿,炎性细胞浸润,血脉通胶囊高、XMT中剂量组心肌总体病变较模型组轻,心肌细胞变性、坏死不明显,肌细胞轻度肿胀、坏死。通过半定量统计分析显示心肌组织血管扩张充血程度、心肌间质水肿程度、心肌变性、坏死程度、炎细胞浸润程度差异具有显著性(P<0.05)。结论: 血脉通胶囊对结扎大鼠冠状动脉致AMI有显著的保护作用。

Abstract:Objective: To explore the pathomorphological changes of myocardial of rats with acute myocardial infarction and the interfering effects of Xuemaitong capsule(XMT). Method: The Wistar rats were randomized into 5 groups, the model group, the sham operation group, the three difference dose groups of XMT preconditioning. The model rats were treated by continuous 14 days intragastric administration. After 14 days, except the sham operation, myocardial infarction of the other groups were established by ligation of coronary artery.Before and after ligation, electrocardiogram were recorded in 5,30,60,90, 120 minutes, 24 hours;pathomorphological change were checked by HE staining. Result: Compared with the high dose, in middle dose and low dose group, the J point were decreased. Pathological alteration of myocardial infarction showed that myocardial endometrial focal disordered arrangement, muscle cells swelling or necrosis, adventitia focal necrosis, and infiltration of inflammatory cells. Compared high and middle groups to model group, the pathomorphological change were slightly among high and middle groups. By semi-quantitative, the pathomorphological changes proved to be true (P<0.05). Conclusion: XMT has important protective function for acute myocardial infarction.
