健脾益气活血汤影响血管紧张素l-7相关信号通路调节自发性高血压大鼠血管活性因子释放机制研究

摘    要:目的: 探讨健脾益气活血汤通过血管紧张素转换酶2(ACE2)、血管紧张素1-7(Ang 1-7)-MAS受体、蛋白激酶B(AKT)信号通路调节自发性高血压大鼠(SHR)血管活性因子释放的作用。方法: 将60只24周龄SHR分为SHR组(给予蒸馏水)、培哚普利组(0.4 mg·kg-1·d-1)、培哚普利联合中药组(培哚普利0.4 mg·kg-1·d-1+健脾益气活血汤20 g·kg-1·d-1,中加西组)、健脾益气活血汤高、中、低剂量组(40,20,10 g·kg-1·d-1),每组10只,以同龄同种系正常血压的京都种大鼠(WKY)10只作为WKY组(给予蒸馏水)。无创血压计监测大鼠尾动脉收缩压;治疗6周后腹主动脉取血测定血浆Ang(1-7)含量,取肾脏提取总RNA及总蛋白,RT-PCR法检测ACE2,MAS mRNA含量表达,Western blot法检测AKT蛋白表达水平。结果: SHR组ACE2,Ang(1-7),MAS,AKT含量与WKY组相比明显减少(P<0.01);培哚普利组、中加西组收缩压与SHR组相比明显降低(P<0.01);培哚普利组、中加西组、中药高剂量组ACE2,Ang(1-7),MAS,AKT含量明显高于SHR组 (P<0.01)。结论: 健脾益气活血汤通过ACE2-Ang (l-7)-MAS-AKT信号通路平衡血管内皮舒缩因子的分泌与释放,发挥改善血管内皮功能及抗高血压作用。

Abstract:Objective: To study the effect of the Chinese medicinethat Jianpi Yiqi Huoxue decoction on angiotensin converting enzyme 2(ACE2)-angiotensin (1-7)Ang(1-7)]-MAS-protein kinase B(AKT)signal pathways in spontaneously hypertensive rats(SHR). Method: Sixty 24-week-old SHRs were randomly divided into six groups:SHR group (distilled water), perindopril group (perindopril 0.4 mg·kg-1·d-1), perindopril plus herb group (perindopril 0.4 mg·kg-1·d-1+10 g·kg-1·d-1of herb), high, medium and low herb group (40,20,10 g·kg-1·d-1). And Wistar kyoto rats(WKY) group. The changes of systolic pressure were measured by BP-98A intelligent sphygmomanometer. After six weeks' treatment, the content of Ang(1-7), ACE2, MAS, AKT was determined. Result: The systolic pressure in the perindopril group and the perindopril plus herb group was prominently decreased, compared with SHR group (P<0.01). The content of Ang(1-7), ACE2, MAS, AKT in the SHR group was lower than those in the WKY group, the perindopril group, the perindopril plus herb group, and the high dose of herb groups(P<0.01). Conclusion: Jianpi Yiqi Huoxue decoction can promote endothelial vasomotor factors secretion through the ACE2-Ang (l-7)-MAS-AKT signaling pathway.
