高效液相色谱法测定复方苦参洗剂中盐酸药根碱、盐酸巴马汀和盐酸小檗碱的含量

摘    要: 目的建立同时测定复方苦参洗剂中盐酸药根碱、盐酸巴马汀和盐酸小檗碱含量的高效液相色谱方法。方法样品以 微孔滤膜过滤后进样。采用Agilent Extend C18色谱柱(4.6 mm×150 mm,5 μm),以水-乙腈(55:45,每100 mL中含磷酸二氢钾 0.34 g,含十二烷基硫酸钠0.17 g)为流动相,紫外检测波长345 nm,柱温为35℃,流速1 mL·min-1。结果在该色谱条件下, 盐酸药根碱、盐酸巴马汀和盐酸小檗碱分别在0.305~6.1,6.4~64,6~60 mg·L-1内呈良好线性关系,r=0.999 1,0.999 5,0.999 6(n=6),平均回收率分别为99.7%,99.8%和100.7%,RSD分别为1.67%,0.87%,0.74%(n=6)。结论 该法可以同时 测定盐酸药根碱、盐酸巴马汀和盐酸小檗碱的含量,检测快速、定量准确,可用于复方苦参洗剂的质量控制。

Abstract:OBJECTIVE To establish a HPLC method for determining the contents of jatrorrhizine hydrochloride(JH), palmatine hy-drochloride(PH) and berberine hydrochloride(BH) in compound Kushen lotion simultaneously.METHODS The separation was performed on an Agilent Extend C18(4.6 mm×150 mm,5 μm) .The mobile phase consisted of water-acetonitrile (45:55,0.34 g potassium dihydrogen phosphate and 0.17 g sodium dodecyl sulfate per 100 mL) with the flow rate of 1.0 mL·min-1. UV detection wavelength was at 345 nm and column temperature was 35 ℃.RESULTS Good linearities were obtained over the range of 0.305~6.1 mg·L-1(r=0.999 1,n=6) for JH, the range of 6.4~64mg·L-1(r=0.9995,n= 6)for PH and the range of 6~60 mg·L-1(r= 0.999 6,n=6) for BH, respectively.The average recoveries of JH, PH and BH were 99.7% (RSD = 1.67% , n=6) ,99.8% (RSD = 0.87% , n= 6)and 100.7% (RSD = 0.74% ,n= 6) , respectively. CONCLUSION This method is rapid and accurate for the simultaneous analysis of JH, PH and BH, and may provide a reliable way for the quality control for compound Kushen lotion.
