人红白血病细胞株c-myc/N-ras/P_(53)mRNA的表达及其与细胞生物学特性的关系

摘    要:本实验用一定浓度的苦参碱作用K562细胞16h, 24h,48h,采用分子生物学Northern Blot和Dot Blot杂交技术分析c-myc,N-ras及P53 mRNA在K562细胞诱导分化过程中表达水平改变,以探讨其表达水平与细胞生物学特性的关系。结果显示苦参碱作用K562细胞24h时,N-ras基因mRNA表达活化;P53-基因表达增强:作用48h时,c-myc mRNA水平表达明显受抑。说明苦参碱在诱导K562细胞分化启动时,能明显改变早期相关癌基因的表达。提示c-myc,N-ras和P53等癌基因是一类重要的细胞增殖、分化调控因素。

Abstract:In this study, c-myc, N-ras and P53 mRNA expression levels were detected respectively by molecular hybridization techniques such as Northern Blot and Dot Blot in K562 cells untreated or treated with Matrine for 16h, 24h, 48h, and the expression levels and cytobiological features were compared. The results showed that N-ras and P53 mRNA expression were markedly increased in K562 cells treated with Marine for 24h; by contrast, for 48h, c- myc mRNA expression was significantly inhibited. It indicated that the expression of the relative oncogene was changed significantly on the start of differentiation induced by Matrine. It suggests that the c-myc, N-ras and P53 on cogenes are one of the key players in the control of cell proliferation and differentiation.
