腺病毒载体介导的色素上皮衍生因子对大鼠视网膜缺血再灌注损伤后视觉电生理的影响

摘    要:目的:观察重组腺病毒介导的色素上皮衍生因子(Ad-PEDF)对大鼠视网膜缺血再灌注损伤后视网膜电流图的影响。方法:选用健康大鼠24只,随机分为正常组、缺血再灌注组、缺血再灌注+Ad-CMV组,缺血再灌注+Ad-PEDF组,以前房加压的方法制备大鼠视网膜缺血再灌注模型,缺血再灌注+Ad-CMV组,缺血再灌注+Ad-PEDF组分别玻璃体腔注射Ad-CMV或Ad-PEDF1μL(滴度3.8×109/PFU),每组按照时间点12,24,72,168h分为4亚组,以ERG分别测量双眼各时期ERGb波、Ops波波幅。结果:缺血再灌注后ERGb及Ops波幅明显降低,与正常组相比在各时间点有明显的统计学差异,缺血再灌注组ERGb波及Ops波幅24h降低最为明显,与缺血12,72,168h相比差异具有显著性。AD-PEDF能够明显促进ERGb波及Ops波幅恢复,与缺血组、缺血再灌注+AD-CMV在各时间点均有明显统计学差异。结论:腺病毒介导的色素上皮衍生因子玻璃体腔注射能够促进大鼠视网膜缺血再灌注损伤功能恢复。

Abstract:AIM: To study the eletroretinograms changes of adenovirus-mediated pigment epithelium-derived factor (Ad-PEDF) on retinal ischemia-reperfusion (RIR) in rats.·METHODS:RIR was induced in 24 adult rats by increasing intraocular pressure via an intracameral infusion. All of the rats were randomly divided into 4 groups: normal control, RIR, RIR+Ad-PEDF treatment and RIR+Ad-CMV control group. RIR+Ad-PEDF treatment and RIR+Ad-CMV control group were undergone intravitreal injection with Ad-PEDF or Ad-CMV 1μL(titer 3.8×109/PFU). Eletroretinograms (ERGs) were performed to determine the functionality of the retinas. Flash ERG results were recorded to assess the retinal function of Ad-PEDF injected eyes and control eyes. ERG studies were performed in both eyes before ischemia (baseline), 12, 24, 72 and 168 hours after reperfusion. The amplitudes of the b-waves and Ops waves were observed.·RESULTS:Beginning from the 12 hours after reperfu-sion, in Ad-CMV control group, the amplitudes of ERG b-wave and Ops wave reduced continuously. Significantly larger amplitudes of ERG b-waves and Ops waves were measured in eyes treated by Ad-PEDF than those in eyes treated by AD-CMV. These results were statistically significant (P<0.05, respectively).·CONCLUSION:The results show that intravitreal injection of Ad-PEDF after rat ischemia-reperfusion injury improves electrophyslology of vision.·
