雌性大鼠在摘除卵巢后延髓内脏带神经元的FOS表达

摘    要:为观察卵巢摘除后“延髓内脏带”内神经元的FOS表达,本文将20只成年雌性大鼠随机分为四组:(1)卵巢摘除+棉籽油组:摘除双侧卵巢后,皮下注射棉籽油(1ml/kg/d);(2)卵巢摘除+雌性激素组:皮下注射苯甲酸雌二醇(10μg/d);(3)卵巢摘除+雄性激素组:皮下注射丙酸睾丸酮(睾因,130μg/d);(4)手术但不摘除卵巢+棉籽油组。所有动物成活6周后处死,取延髓切片进行抗FOS蛋白和抗酪氨酸羟化酶的免疫组化反应。结果在第4组的“延髓内脏带”出现许多FOS阳性神经元,其中40%为FOS/酪氨酸羟化酶双重反应阳性;第2组的FOS阳性神经元次之;第3组则更少;最少者为第1组。说明“延髓内脏带”内神经元(特别是酪氨酸羟化酶阳性神经元)对体内性激素的变化起反应。

Abstract:In order to study c-fos expression in neurons of medullary visceral zone of female rat after ovariectomy, 20 adult female rats (200 g b. w) were divided into four groups. (1) first group (O+C group) rats. ovariectomized, followed by subcutanous injection of cottonseed oil (1 ml/kg b. w/day). (2) Second group (O+E group) rats, ovariectomized, followed by subcutanous injection of estradiol benzoate (10 μg/day). (3) Third group (O+T group) rats, ovariectomized, followed by subcutanous injection of testosterone propionate (130 μg/day). (4) Fourth group (S+C group) rats, not ovariectomized, followed by subcutanous injection of cottonseed oil. After 6 weeks. all animals were sacrificed, and the medulla oblongata were removed.The sections of the medulla oblongata were subjected to anti-FOS and anti-tyrosine hydroxylase (TH) immunohistochemical reaction with ABC method. The results of the experiments were that many FOS-positive neurons were found in MVZ of S+C group rats, 40% of them were FOS/TH double labeled ones, number of FOS positive neurons in O+E group rats were next to it, FOS positive neurons appeared in O+T group were small in number. Smallest number of FOS positive neurons were found in O+C group. These results suggest that the neurons (specially TH neurons) in MVZ respond on change of sex hormone.
