应用不同方法检测P糖蛋白在肺癌组织中的表达

[目的]探讨应用不同方法检测P糖蛋白(P - glycoprotein ,Pgp)在不同人群下不同组织类型和分化程度肺癌组织中的表达情况。[方法]收集肺癌组织标本4 16例,显微镜下观察肿瘤的组织类型和分化情况,将肺癌组织按照病理切片情况分组,然后分别用免疫组织化学法、RT -PCR和流式细胞仪检测术检测P糖蛋白在肺癌组织中的表达情况。[结果]免疫组织化学法、RT -PCR和流式细胞仪检测术检测阳性结果分别是37%、5 8%和5 2 %。Pgp阳性结果在高年龄组和复诊组较其他组别高(P <0 .0 5 )。在不同组织类型和分化程度肺癌组织中无差别。[结论]在同免疫组织化学法比较上看,RT -PCR和流式细胞仪检测术方法简单、快速、灵敏和特异,可用于临床检测肺癌组织中P糖蛋白。P糖蛋白在肺癌组织中的表达程度与年龄和初/复诊等情况有关而与组织类型和分化程度无关。

To analyses expression of Pgp in lung neoplasms with different method

ObjectiveTo try to analyze the expression of Pgp in lung neoplasms by three methods.[Method]To obtained 416 surgical biopsy specimens, the expression of Pgp in lung neoplasms was determined by IH,FC and RT-PCR.[Results]The positive result were 37%, 58% and 52% by IH,CF and RT-PCR methods. The positive rate of Pgp expression was significantly higher in the elden group and re-diagnosis group than the other group (P<0.05). There is not significant difference in pulmonany pathological score. [Conclusion]The method of FC and RT-PCR were simple;rapid;sensitive and specific;and could be used for detecting Pgp in clinic.The expression of Pgp in lung neoplasms were significantly associated with ages and second treatment ;and were not significantly associated with lung pathological sore.
