摘    要:目的利用PCR-SSCP银染法检测不同吸烟时间段的大鼠肺组织p53、K-ras基因突变情况。方法建立Wistar大鼠被动吸烟模型。利用PCR-SSCP法检测p53第5 ̄8外显子、K-ras第1外显子在吸烟各阶段的突变情况。结果p53基因突变率随吸烟时间的延长而增加(P<0.05),并呈逐渐上升趋势,K-ras基因随吸烟时间的延长突变率增加不显著。结论在吸烟所致肺癌的发生发展中p53基因的突变是早期事件,为通过筛选吸烟者痰液中脱落细胞中的p53基因突变进行肺癌的早期诊断提供了理论依据。

Abstract:Objective To observe the mutation of p53 and K-ras gene in the lung of smoking rats at the different time by PCR-SSCP. Methods The model of smoking wistar rat was used to examine the mutation in the exon 5, 6, 7-8 of p53 and the exon 1 of K-ras by PCR-SSCP. Results The mutation rate of p53 gene was increased as the smoking time prolonged(P<0.05 ), this phenomenon was trending to rise,but not significant for K-ras gene. Conclusion p53 gene mutation was early event in the carcinogenesis and development of rat lung carcinoma induced by smoking,which provided a theoretical evidence for the early diagnosis of smoking-related lung carcinomas through screening p53 gene mutation of cast-off cells in smoker's sputum.
