胸苷激酶和胞嘧啶脱氨酶融合基因抑骨肉瘤生长的研究

摘    要:目的观察单纯疱疹病毒-胸苷激酶/胞嘧啶脱氨酶融合自杀基因(HSV-TK/CD)对裸鼠骨肉瘤生长的影响。方法人骨肉瘤细胞系MG-63细胞于裸鼠成瘤后将其分为6组:(1)PBS对照组;(2)重组腺病毒(adenovirus,Ad)-Lac-Z/GCV 5-FC组;(3)Ad-TK/GCV组;(4)Ad-CD/5- FC组;(5)Ad-TK/GCV Ad-CD15-FC组;(6)Ad-TK-CD/GCV 5-FC组。各组经治疗后观察肿瘤的生长状况及组织病理学改变。结果(2)组与(1)组比较,肿瘤体积与肿瘤坏死程度差异无统计学意义(P>0.05);与(1)、(2)组比较,(3)~(6)组肿瘤的生长均受到明显抑制(P<0.05),且(5)、(6)组与(3)、(4)比较差异有统计学意义(P<0.05)。结论融合自杀基因HSV-TK/CD对裸鼠骨肉瘤生长有明显的抑制作用。

Abstract:Objective To observe the effect of herpes simplex virus-thymidine kinase/cytosine deaminase fusion suicide gene on the osteosarcoma growth of nude mice. Methods Osteosarcoma model of nude mouse was made with MG-63 cells injected into axillary fossa. Forty -eight nude mice were divided randomly into 6 groups: (1) PBS control group; (2) Ad-Lac-Z/GCV 5-FC group; (3) Ad-TK/GCV group; (4) Ad-CD/5-FC group; (5) Ad-TK/GCV Ad-CD/5-FC group; (6) Ad-TK-CD/GCV 5-FC group. Tumor volume, inhibitory rate of tumor growth and histopathology were analyzed after each group was treated with gene and prodrug. Results There was no significant difference in tumor volume and rate of tumor cell death between Ad-Lac-Z/GCV 5-FC group and PBS control group. Tumor growth in Ad-TK/CCV group, Ad-CD/5-FC group, Ad-TK/GCV Ad-CD/5-FC group and Ad-TK-CD/GCV 5-FC group was inhibited significantly as compared with PBS control group and Ad-Lac-Z/GCV 5-FC group (P <0.05). There was significant difference between Ad-TK/GCV Ad-CD/5-FC group, Ad-TK-CD/GCV 5-FC and Ad-TK/GCV group, Ad-CD/5-FC group (P <0.05). Conclusion HSV-TK/CD fusion suicide gene could significantly inhibit the osteosarcoma of nuce mouse.
