高效液相色谱法测定盆炎灵合剂中的阿魏酸、延胡索乙素、芍药苷、丹酚酸B及原儿茶醛含量

摘    要:目的采用高效液相色谱法同时测定盆炎灵合剂中阿魏酸、延胡索乙素、芍药苷、丹酚酸B和原儿茶醛等成分的含量。方法 Hypersil ODS2色谱柱(4.6 mm×250 mm,5μm);流速:1.0 m L·min-1;流动相A为乙腈,流动相B为0.1%磷酸溶液,梯度洗脱。结果阿魏酸、延胡索乙素、芍药苷、丹酚酸B和原儿茶醛质量浓度分别在0.008 40.168 4 mg·m L-1(r=0.999 9)、0.023 60.168 4 mg·m L-1(r=0.999 9)、0.023 60.471 2 mg·m L-1(r=0.999 3)、0.030 40.471 2 mg·m L-1(r=0.999 3)、0.030 40.304 0 mg·m L-1(r=0.999 7)、0.031 30.304 0 mg·m L-1(r=0.999 7)、0.031 30.625 4 mg·m L-1(r=0.999 9)、0.010 10.625 4 mg·m L-1(r=0.999 9)、0.010 10.202 8 mg·m L-1(r=0.999 9)与其峰面积呈良好的线性关系;平均加样回收率分别为103.9%、96.5%、96.8%、98.7%和101.5%,RSD值分别为1.7%、2.0%、2.5%、2.0%和2.2%(n=6)。结论该方法线性、重复性、精密度、加样回收率均符合要求,可作为盆炎灵合剂的含量控制方法。

Abstract:Objective To develop an HPLC method to determine the content of ferulic acid, tetrahydropalmatine, paeoniflorin, salvianolic acid B and protocatechualdehyde in Penyanling mixture. Methods A Hypersil ODS2(4.6 mm×250 mm,5 μm) column was used and the flow rate was 1.0 m L·min^-1. The mobile phase was acetonitrile and 0.1%phosphoric acid solution with gradient elution. Results Ferulic acid, tetrahydropalmatine, paeoniflorin, salvianolic acid B and protocatechualdehyde had a good linearity at 0.008 4- 0.168 4(r = 0.999 9), 0.023 6- 0.471 2(r = 0.999 3), 0.030 4- 0.304 0(r = 0.999 7), 0.031 3- 0.625 4(r = 0.999 9) and 0.010 1- 0.202 8 mg·mL^-1(r = 0.999 9), respectively. The average recoveries and RSDs were 103.9%(1.7%), 96.5%(2.0%), 96.8%(2.5%), 98.7%(2.0%) and 101.5%(2.2%), respectively. Conclusion The method is stable, precise and repeatable, which can be used to determine the content of ferulic acid, tetrahydropalmatine, paeoniflorin, salvianolic acid B and protocatechualdehyde in Penyanling mixture.
