肝硬化患者外周血IL-8及其mRNA表达与HBV载量相关性

摘    要:目的 探讨肝硬化患者血清IL-8含量、外周血单个核细胞(PBMCs)中IL-8 mRNA表达及其与HBV载量相关性。方法:以ELISA法检测65例肝硬化患者(代偿期36例,失代偿期29例)血清IL-8含量,定量PCR检测患者外周血IL-8 mRNA水平,设GAPDH为参照,以logcDNA/logGAPDH比值代表其最终mRNA水平。以Real-time PCR法检测患者外周血HBV载量。结果:肝硬化患者外周血IL-8及其mRNA表达量分别为(448.41±59.46)pg/ml、(1.4850±0.1801)logcDNA/logGAPDH,较正常组显著增高,差异有显著性(P<0.01)。其中,失代偿组IL-8及其mRNA分别为(488.19±49.44)pg/ml、(1.5453±0.1783)logcDNA/logGAPDH,较代偿组升高,差异有显著性(P<0.01, P<0.05)。病毒复制组外周血IL-8及其 mRNA水平分别为(472.21±52.44)pg/ml、(1.5676±0.1558) logcDNA/logGAPDH,与HBV载量相关系数分别为0.487、0.454,较非复制组显著升高,差异有统计学意义(P<0.01)。结论:肝硬化外周血IL-8及其 mRNA表达升高,不仅与体内病毒复制水平密切相关,且与患者病情严重程度有关。IL-8在肝硬化的致炎细胞分子病理损伤过程中起重要作用。

Expression of IL-8, IL-8 mRNA and correlation with loads of HBV in the peripheral blood of cirrhosis

Abstract:Objective: To study the expression of IL-8 in serum and IL-8 mRNA in peripheral blood mononuclear cells (PBMCs) and the correlation with loads of HBV in the cirrhosis. Methods: The levels of IL-8 in serum of sixty-five cirrhosis (compensated cirrhosis 36, decompensated cirrhosis 29) were detected by ELISA, and the expression of IL-8 mRNA in the peripheral blood of cirrhosis were simultaneously explored by the quantitative PCR. The ratio of logcDNA/logGAPDH was regarded as the extreme level of IL-8 mRNA. The loads of HBV in PBMCs and serum were detected by Real-time PCR. Results: The levels of IL-8 and its mRNA in peripheral blood of the patients were (448.41±59.46) pg/ml, (1.4850±0.1801) logcDNA/logGAPDH, which were both significantly higher than those in normal controls (P<0.01). The high levels of IL-8 (488.19±49.44)pg/ml) and its mRNA (1.5453±0.1783) logcDNA/logGAPDH both had been identified higher in decompensated cirrhosis compared with the compensated cirrhosis (P<0.01). They were also significantly higher in the group with positive of HBV-DNA (472.21±52.41 pg/ml, 1.5676±0.1558 logcDNA/logGAPDH) than those in the group with negative of HBV-DNA (P<0.01). The correlation coefficients between CXCR1, CXCR2 and the loads of HBV were 0.487, 0.454. Conclusions: The high levels of IL-8 and its mRNA expression are confirmed in cirrhosis and have significantly associated with the loads of HBV, and also related with the disease progression of cirrhosis. IL-8, a key type of inflammatory cytokine or chemokine, may play an important role in the cellular and molecular mechanisms of pathological damage in cirrhosis.
