金丝桃苷预处理对大鼠全脑缺血再灌注后神经行为学的影响

摘    要:目的探讨金丝桃苷预处理对大鼠全脑缺血再灌注后神经行为学的影响及其可能机制。方法60只SD雄性大鼠随机分为假手术组、模型组及金丝桃苷预处理组各20只。金丝桃苷预处理组每日腹腔注射金丝桃苷连续7 d。行四血管阻断全脑缺血10 min再灌注,再灌注后24 h每组各取5只,右侧大脑采用干湿重法计算脑水含量,左侧大脑取海马测定SOD活性及MDA含量。其余每组各15只大鼠分别于再灌注前、再灌注后6 h1、2 h、24h4、8 h、96 h进行改良NSS评分;再灌注后第5天开始进行Morris水迷宫实验。结果模型组脑水含量、SOD活性、MDA含量、NSS评分、平均潜伏期及探索实验中在第2象限时间比与假手术组比较有显著性差异(P均<0.05);金丝桃苷预处理组与模型组比较,脑水含量降低,SOD活性增高,MDA含量降低,NSS评分降低,平均潜伏期缩短,第2象限时间比增高(P<0.05)。结论金丝桃苷可减轻大鼠全脑缺血再灌注后脑水肿程度,缓解海马区自由基代谢异常,改善认知功能。

Abstract:Objective It is to approach the influence of hyperin preconditioning on neuroethology in rats with global cerebral ischemia reperfusion injury so as to investigate its probable mechanism.Methods 60 SD male rats were randomly divided into sham operated group,model group and hyperin preconditioning group.The animals in hyperin preconditioning group were intraperitoneally administered with hyperin for 7 days.The operation of global cerebral ischemia reperfusion injury was executed by pullsinelli-4 vessel occlusion.5 rats in each group were killed to obtain their brains after 24 h to successful modeling.The water content in the right brain was measured by dry-wet method,and the activity of SOD and the content of MDA in the left brain were measured.Then neurological severity scores(NSS) of the other rats in each group were observed respectively before reperfusion and after 6 h,12 h,24 h,48 h,96 h.The spatial learning and memory were tested respectively by Morris water maze test in the 5th day after to reperfusion.Results The above mentioned indexes in the model group showed differences compared with those in the sham operated group(P<0.05).Compared to the model group,the water content and the content of MDA in the brain tissue,the NSS and the mean latency were decreased,and the activity of SOD and the percent of time course in the second quadrant increased in hyperin preconditioning group(allP<0.05).Conclusion Hyperin can reduce the cerebral edema degree,relieve the abnormity of free radical,and exert protective effect on cognitive impairment in global cerebral ischemia reperfusion injury rats.
