复方黄连胶囊对早期糖尿病肾病大鼠肾组织VEGF蛋白和mRNA表达的影响研究

摘    要:目的:探讨复方黄连胶囊(CRCC)对早期糖尿病肾病大鼠肾组织VEGF蛋白和mRNA表达的影响。方法:腹腔一次性注射链脲佐菌素复制早期糖尿病肾病大鼠模型,分为正常对照、模型、消渴丸(0.8g·kg-1)及CRCC低、高剂量(生药含量分别为2.18、4.36g·kg-1)组,灌胃给药每日1次,8周后检测大鼠空腹血糖(FBG)、肌酐、尿素氮;放免分析检测血清胰岛素(Ins)、α1微球蛋白(α1-MG)、β2微球蛋白(β2-MG)水平;免疫组化技术和逆转录聚合酶链反应(RT-PCR)检测肾组织VEGF蛋白和mR-NA表达。结果:与模型组比较,CRCC给药组大鼠FBG、α1-MG、β2-MG显著降低(P<0.01),Ins显著升高(P<0.01),VEGF蛋白和mRNA的表达减弱(P<0.05)。结论:CRCC可显著降低早期糖尿病肾病大鼠肾组织VEGF蛋白和mRNA的过度表达,改善肾功能,延缓糖尿病肾病发展。

Abstract:OBJECTIVE: To explore the effects of compound Rhizoma coptidis capsule on the expressions of vascular endothelial growth factor (VEGF) and mRNA in the kidney of rats with early diabetic nephropathy. METHODS: The rats were intraperitoneally administered with streptozotocin to establish early diabetic nephropathy model. Then the rats were randomly divided into five groups: normal control group, diabetic nephropathy model group, Xiaoke Pill (0.8 g·kg^-1) group, CRCC-treated groups (2.18 g ·kg ^-1 and 4.36 g·kg^-1) . The animals were intragastrically administered with corresponding drugs once daily. After 8 weeks, the fasting blood glucose, blood urea nitrogen and creatinine werc measured and serum levels of insulin (Ins), a1-MG, β2 MG were determined by radioimmunoassay. Immunohistochemistry and RT PCR were used to detect the expressions of VEGF and mRNA in the renal tissue of rats. RESULTS: In CRCC treated groups, serum levels of FBG, a1- MG,β2-MG, BUN and Scr were markedly decreased (P 〈 0.01), while the insulin level increased markedly(P 〈 0.01), and the expressions of VEGF and mRNA in renal glomeruli and tubuli were decreased(P 〈0.05) .CONCLUSION: CRCC can obviously improve renal function and decrease the overexpression of VEGF and mRNA in kidney tissue, and delay the progression of diabetic nephropathy in diabetic nephropathy model rats.
