姜黄素对血管瘤内皮细胞凋亡的诱导作用及对Bcl-2和Bax蛋白表达的调控

摘    要:目的:研究姜黄素对血管瘤内皮细胞凋亡的影响。方法:在培养液体系中加入不同浓度的姜黄素进行不同时间长度的诱导,以MTT法检测细胞增殖,流式细胞术,TUNEL法检测细胞凋亡情况,Western blotting分析Bcl-2和Caspase-9蛋白表达。结果:1~50μmol/姜黄素均能抑制血管瘤内皮细胞增殖。5、10、50μmol/L姜黄素作用细胞4天后的凋亡率分别为(11.98±2.13)%、(17.96±1.25)%、(26.86±2.08)%,与对照组(4.03±1.16)%比较差异均有显著性意义(均P<0.05)。姜黄素浓度越高,诱导凋亡作用越明显,呈明显的剂量依赖效应。50μmol/L姜黄素作用细胞1、2、3、4天后的凋亡率分别为(6.93±1.06)%、(10.15±1.35)%、(17.21±2.06)%、(26.83±2.02)%,与对照组(3.98±1.03)%比较差异均有显著性意义(均P<0.05)。姜黄素作用时间越长,抑制血管瘤内皮细胞增殖作用和诱导凋亡的作用也越明显,呈明显的时间依赖效应。同时姜黄素浓度越大、作用时间越长,血管瘤内皮细胞细胞中Bcl-2蛋白的表达明显减少,而Caspase-9蛋白的表达明显增加。结论:姜黄素能够抑制血管瘤内皮细胞增殖、诱导血管瘤内皮细胞发生凋亡,并呈时间和剂量依赖效应,可能是通过下调凋亡抑制基因bcl-2表达,上调Caspase-9表达而实现的。

Influence of Curcumin on Inducing Endothelial Cells Apoptosis of Angioma And Regualting Expressions of Bcl-2 And Bax

Abstract:Objective:To investigate the effects of curcumin on inducing apoptosis of endothelial cells of angioma.Methods:Different concent rations of curcumin were added into culture media system to induce apoptosis of endothelial cells.The apoptosis was detected by MTT assay,flow cytometry and TUNEL method.Results:Curcumin at concent rations of 1-50μmol/L could inhibit the proliferation.After the cells were treated with 5,10 and 50μmol/L curcumin for 4 days,the rates of apoptosis were(12.46±2.37)%,(18.36±1.32)% and(27.15±2.17)% respectively,showing significant difference with that in control group(4.09±1.18)%,P<0.05.The rates of apoptosis induced by 50μmol/L curcumin for 1,2,3 and 4 days were(7.35±1.28)%,(10.72±1.68)%,(17.65±2.14)% and(27.15±2.17)% respectively,significantly higher than in control group(4.09±1.18)%,P<0.05,showing a time-dependent manner.At the same time the concentration is more big and act time more longer,the expressions of Bcl-2s obviously reduced,while the expression of Caspase-9s obviously.Conclusion:Curcumin could induce apoptosis in a time and dose-dependent manner,which may be an approval to decrease expressions of bcl-2,and increade Caspase-9.
